Alzheimer’s disease (AD) is the leading dementia among elderly with complex origins. Despite extensive investigation into AD-associated protein-coding genes, involvement of noncoding RNAs (ncRNAs) and posttranscriptional modification (PTM) in AD pathogenesis remains unclear. Here, we comprehensively characterized landscape ncRNAs PTM events 1460 samples across six brain regions sourced from Mount Sinai/JJ Peters VA Medical Center Brain Bank Study Mayo cohorts, encompassing 33,321 long...
NOD2 functions as an intracellular sensor for microbial pathogen and plays important role in epithelial defense. The loss-of-function mutation of is strongly associated with human Crohn's disease (CD). However, the mechanisms how maintains intestinal homeostasis regulates susceptibility CD are still unclear. Here we found that numbers intraepithelial lymphocytes (IELs) were reduced significantly Nod2(-/-) mice residual IELs displayed proliferation increased apoptosis. Further study showed...
Clonal hematopoiesis (CH) is a phenomenon caused by expansion of white blood cells descended from single hematopoietic stem cell. While CH can be associated with leukemia and some solid tumors, the relationship between lung cancer remains largely unknown. To help clarify this relationship, we analyzed whole-exome sequencing (WES) data 1,958 cases controls. Potential mutations were identified set hierarchical filtering criteria in different exonic regions, associations number clinical traits...
Abstract Recent studies suggest that rare variants exhibit stronger effect sizes and might play a crucial role in the etiology of lung cancers (LC). Whole exome plus targeted sequencing germline DNA was performed on 1045 LC cases 885 controls discovery set. To unveil inherited causal variants, we focused predicted deleterious small indels enriched or controls. Promising candidates were further validated series 26,803 LCs 555,107 During discovery, identified 25 associated with susceptibility,...
Highlights•We explored identified specific markers of M1 and M2 macrophage.•The potential therapeutic compounds with were explored.•Discovering these promotes an understanding mechanisms OC.AbstractPurposeWe aimed to analyze the sensitivity patients chemotherapy drugs actively explore new intervention targets, providing essential reference for personalized treatment.MethodsCandidate significant differential expression in macrophages by analyzing gene at single-cell level. A weighted...
Abstract Multiple epigenetic changes, including alterations in DNA methylation occur during tumorigenesis. Various inhibitors of have been developed to prevent proliferation cancer cells. 5-fluoro-2 ′ -deoxycytidine (FCdR) is one such inhibitor, which currently phase II clinical trial. To investigate the molecular mechanism/s by FCdR might mediate repression tumor cell proliferation, we analyzed toxicity various lines established from different sarcomas. We found HCT116, a colon line, much...
Tail-cut bovine spermatozoa were microinjected into ooplasmic lipid polarised, in vitro matured oocytes using a piezomicropipette-driving system. No exogenous oocyte activation treatment was used. Of the sperm-injected oocytes, 86.3% activated, 71.8% cleaved and 22.7% developed to blastocyst stage. The average cell count of blastocysts 122.5 +/- 15 majority (81.8%) cytologically normal (diploid). When transferred recipient cows, 5 8 fetuses 4 7 recipients became pregnant. Normal offspring born.
This study was conducted to investigate the possibility of using bovine oocytes for a heterologous fertility test by intracytoplasmic sperm injection (ICSI) and compare pronuclear formation ram, bull minke whale spermatozoa after into oocytes. Bovine were cultured in vitro 24 h those with polar body selected ICSI. Frozen-thawed semen from three species treated 5 mM dithiothreitol 1 killed storing them -20 °C refrigerator before use. ICSI performed Piezo system. Three experiments designed. In...
This study was conducted to investigate the effects of ooplasm clarification, sperm tail-cutting and reduction polyvinylpyrrolidone (PVP) concentration in injection vehicle solution on ICSI cattle. In vitro matured oocytes, motile spermatozoa a piezo system were used for ICSI. experiment 1, oocytes centrifuged at 6,000 × g 7 min polarize lipid before Sperm carried out effect tested 2. 3, PVP investigated. After ICSI, cultured mTALP 15 h fixation observation. It found that clarification...
Serous ovarian cancer is one of the major causes related death among women worldwide. The advanced diagnosis worsens prognosis patients with serous cancer. immune system has an important impact on progression Herein, we aimed to establish prognostic signature assist in early diagnosis, treatment, and evaluation Multiple public data sets genes were obtained from various online databases, signatures developed through differential expression analysis, univariate Cox proportional hazard...
Abstract To investigate the association between clonal haematopoiesis (CH) and lung cancer risk, we identified CH mutations in 1 059 cases 899 controls using blood whole‐exome sequencing data generated from Integrative Analysis of Lung Cancer Etiology Risk project International Consortium (INTEGRAL‐ILCCO). Based on variant allele frequency (VAF) these mutations, stratified carriers into two groups, low VAF (1%–10%) high (≥10%), respectively. We observed a significant presence risk after...
Accumulating evidence suggests that the tumor immune microenvironment (TIME) significantly influences response to immunotherapy, yet this complex relationship remains elusive. To address issue, we developed TimiGP-Response (TIME Illustration based on Gene Pairing designed for immunotherapy Response), a computational framework leveraging single-cell and bulk transcriptomic data, along with information, construct cell-cell interaction networks associated responders estimate role of cells in...
Neoantigens are presented on the cancer cell surface by peptide-restricted human leukocyte antigen (HLA) proteins and can subsequently activate cognate T cells. It has been hypothesized that observed somatic mutations in tumors shaped immunosurveillance. We investigated all identified The Cancer Genome Atlas (TCGA) Skin Cutaneous Melanoma (SKCM) samples. By applying a computational algorithm, we calculated binding affinity of resulting neo-peptides their corresponding wild-type peptides with...
Introduction The efficacy of adjuvant chloroquine for glioblastoma remains controversial. We conduct a systematic review and meta-analysis to explore the influence on treatment recurrent glioblastoma. Methods search PubMed, Embase, Web science, EBSCO, Cochrane library databases through January 2020 randomized controlled trials (RCTs) assessing This is performed using random-effect model. Results Three RCTs are included in meta-analysis. Overall, compared with control group glioblastoma,...
Abstract Chromatin accessibility is essential for transcriptional activation of genomic regions. It well established that transcription factors (TFs) and histone modifications (HMs) play critical roles in chromatin regulation. However, there a lack studies quantify these relationships. Here we constructed two-layer model to predict by integrating DNA sequence, TF binding, HM signals. By applying the two human cell lines (GM12878 HepG2), found sequences had limited power prediction, while...
Prophylaxis of brain injury in newborns has been a main concern since the first neonatal neuronal intensive care unit (NNICU) was established world 2008. The aim this study to outline and evaluate unit’s development by analyzing demographics patients, services delivered, short-term outcomes before after establishment NNICU. During two investigation periods, 384 were diagnosed or suspected as “neonatal encephalopathy”, among which 185 patients admitted NNICU between 2011.03.01 2012.09.30...
