Eukaryotic translation initiation factor eIF5A has been implicated in HIV-1 replication. This protein contains the apparently unique amino acid hypusine that is formed by post-translational modification of a lysine residue catalyzed deoxyhypusine synthase and hydroxylase (DOHH). DOHH activity inhibited two clinically used drugs, topical fungicide ciclopirox systemic medicinal iron chelator deferiprone. Deferiprone reported to inhibit replication tissue culture.Ciclopirox deferiprone blocked...

The first ZnO-NP catalyzed Ugi type three-component (AB2C) reaction has been developed for the synthesis of 2-arylamino-2-phenylacetimidamide from an aldehyde, amine and isocyanide in aqueous media. This nanoparticle catalysed (NPCR) is high yielding good atom economy as well efficiency. synthesized phenylacetimidamide yielded 2-amino-2-phenylacetamide on hydrolysis with I2–SDS–water, whereas alkaline afforded N-substituted α-amino acid.

An efficient micelle promoted one-pot synthesis of 3-amino alkylated indoles has been developed via a three-component Mannich-type reaction from secondary amines, aldehydes and in water under mild conditions. In this reaction, micelles stabilize iminium ions, which undergo nucleophilic addition the indole to give very good yields.

HIV-1 blocks apoptosis, programmed cell death, an innate defense of cells against viral invasion. However, apoptosis can be selectively reactivated in HIV-infected by chemical agents that interfere with gene expression. We studied two globally used medicines, the topical antifungal ciclopirox and iron chelator deferiprone, for their effect on H9 peripheral blood mononuclear infected clinical isolates. Both medicines activated preferentially cells, suggesting drugs mediate escape from...

Antiretrovirals suppress HIV-1 production yet spare the sites of production, DNA-harboring cells that evade immune detection and enable viral resistance on-drug rebound off-drug. Therapeutic ablation pathogenic markedly improves outcome many diseases. We extend this strategy to infection. Using drug-based lead discovery, we report concentration threshold-dependent antiretroviral action medicinal chelator deferiprone validate preclinical findings by a proof-of-concept double-blind trial.
