サブスタンスP(SP)に代表されるタキキニン・ペプチド類には,NK-1,NK-2およびNK-3の3つの受容体が存在し種々の生理反応に関与している。中でもSPとNK-1受容体は延髄の最後野や弧束核に密に存在し,嘔吐に強く関与していると考えられるが,詳細な作用機序は明らかではない。そこで本研究では,嘔吐発現機構における特に延髄最後野のタキキニンNK-1受容体に注目し,新たに開発されたNK-1受容体拮抗薬HSP-117による制吐作用についてフェレットを用いて検討した。フェレットの延髄の3H-SPの結合部位は,弧束核,舌下核,迷走神経背側核,および最後野に密に分布していた。さらに最後野を含む延髄切片をSPで灌流すると,最後野の約50%の細胞に興奮反応が認められた。このSPによる神経活動興奮反応はNK-1受容体アンタゴニストのCP-99,994によって著しく抑制されたことから,フェレットの延髄最後野にNK-1受容体が存在することが明らかとなった。タキキニンNK-1受容体拮抗薬HSP-ll7とCP-99,994のNK-1受容体に対する選択性を,IM-9細胞の膜標品に対する3H-SPの結合阻害...
