gp130 is the shared signal-transducing receptor subunit for large and important family of interleukin 6-like cytokines. Previous x-ray structures ligand-receptor complexes this lack three membrane-proximal domains that are essential signal transduction. Here we report crystal structure entire extracellular portion human (domains 1-6, D1-D6) at 3.6 A resolution, in an unliganded form, as well a higher resolution fibronectin type III (D4-D6) 1.9 A. This represents first atomic complete...

The nucleolar surveillance pathway monitors integrity and responds to stress by mediating binding of ribosomal proteins MDM2, resulting in p53 accumulation. Inappropriate activation is implicated the pathogenesis ribosomopathies, while drugs selectively activating are trials for cancer. Despite this, molecular mechanism(s) regulating this process poorly understood. Using genome-wide loss-of-function screens, we demonstrate ribosome biogenesis axis as most potent class genes whose disruption...

The LIM kinase family includes two proteins: LIMK1 and LIMK2. These proteins have identical genomic structure overall amino acid identity of 50%. Both regulate actin polymerization via phosphorylation inactivation the depolymerizing factors ADF/cofilin. Although function endogenous is well established, little known about LIMK2 protein. To understand specific role protein, we examined its expression in embryonic adult mice using a rat monoclonal antibody, which recognizes specifically PDZ...

Dendritic cells (DCs) are heterogeneous, comprising subsets with functional specializations that play distinct roles in immunity as well immunopathology. We investigated the molecular control of cell survival two main DC subsets: plasmacytoid DCs (pDCs) and conventional (cDCs) their dependence on individual antiapoptotic BCL-2 family members. Compared cDCs, pDCs had higher expression BCL-2, lower A1, similar levels MCL-1 BCL-XL. Transgenic overexpression increased pDC pool size vivo only...

Abstract Plasmacytoid dendritic cells (pDCs) play an important role in immunity to certain pathogens and immunopathology some autoimmune diseases. They are thought have a longer lifespan than conventional DCs (cDCs), largely based on slower rate of BrdU labeling by splenic pDCs. Here we demonstrated that pDC expansion therefore pDCs occurs bone marrow (BM). The was similar between BM spleen cDCs. Therefore, likely reflects the “migration time” (∼2 days) for labeled traffic spleen, not...

Female-controlled contraception/HIV prevention is critical to address health issues associated with gender inequality. Therefore, a contraceptive which can be administered in tandem microbicide inhibit sexually transmitted infections, desirable. We aimed determine whether PEGylated LIF inhibitor (PEGLA) was an effective following vaginal delivery identify non-uterine targets of PEGLA...

We have expressed and purified three soluble fragments of the human LRIG1-ECD (extracellular domain): LRIG1-LRR (leucine-rich repeat) domain, LRIG1-3Ig (immunoglobulin-like) LRIG1-LRR-1Ig fragment using baculovirus vectors in insect cells. The two LRIG1 domains crystallised so that we been able to determine three-dimensional structures at 2.3Å resolution. developed a structure for homology modelling based on LINGO-1 structure. domain are monomers solution, whereas appears be dimeric. could...

The nucleolus is a dynamic subnuclear compartment that has number of different functions, but its primary role to coordinate the production and assembly ribosomes. For well over 100 years, pathologists have used changes in nucleolar size stage diseases such as cancer. Abstract The nucleolar surveillance pathway (NSP) monitors fidelity and responds to stresses (i.e., inactivation of ribosome biogenesis) by mediating the inhibitory binding ribosomal proteins (RPs) mouse double minute 2 homolog (MDM2), a nuclear-localised E3 ubiquitin ligase, which results in p53 accumulation. Inappropriate activation NSP has been implicated pathogenesis collection human diseases termed “ribosomopathies”, while drugs that selectively activate are now trials for cancer....
