Platinum-based anticancer drugs cause neurotoxicity. In particular, oxaliplatin produces early-developing, painful, and cold-exacerbated paresthesias. However, the mechanism underlying these bothersome dose-limiting adverse effects is unknown. We hypothesized that transient receptor potential ankyrin 1 (TRPA1), a cation channel activated by oxidative stress cold temperature, contributes to mechanical hypersensitivity caused cisplatin. Oxaliplatin cisplatin evoked glutathione-sensitive...

Despite intense investigation, the mechanisms of different forms trigeminal neuropathic pain remain substantially unidentified. The transient receptor potential ankyrin 1 channel (encoded by TRPA1 ) has been reported to contribute allodynia or hyperalgesia in some models, including those produced sciatic nerve constriction. However, role and processes that cause pain-like behaviours from insult are poorly understood. TRPA1, monocytes macrophages, oxidative stress behaviour evoked...

The California bay laurel or Umbellularia californica (Hook. & Arn.) Nutt., is known as the 'headache tree' because inhalation of its vapours can cause severe headache crises. However, underlying mechanism precipitating properties unknown. monoterpene ketone umbellulone, major volatile constituent leaves californica, has irritating properties, and a reactive molecule that rapidly binds thiols. Thus, we hypothesized umbellulone stimulates transient receptor potential ankyrin 1 channel in...

Summary Parthenolide, a major constituent of feverfew, acts as partial agonist TRPA1. Parthenolide’s ability to target TRPA1 could explain its therapeutic effects on migraine. Although feverfew has been used for centuries treat pain and headaches is recommended migraine treatment, the mechanism protective action remains unknown. Migraine triggered by calcitonin gene-related peptide (CGRP) release from trigeminal neurons. Peptidergic sensory neurons express series transient receptor potential...

Ethanol stimulating transient receptor potential vanilloid 1 (TRPV1) on primary sensory neurons promotes neurogenic inflammation, including calcitonin gene-related peptide (CGRP)-mediated coronary dilation. Alcoholic beverages trigger migraine attacks and activation of trigeminal plays a role in migraine. We have investigated guinea pigs whether ethanol by TRPV1 stimulation causes inflammation the trigeminovascular system. Ethanolevoked release neuropeptides from slices dura mater was...

In our pursuit to identify selective ligands for Bz/GABAA receptor subtypes, a novel pyrazolo[1,5-a]pyrimidine derivative (4), the azaisostere of zolpidem, was synthesized and evaluated in vitro on bovine brain homogenate recombinant benzodiazepine receptors (αxβ2/3γ2, x = 1−3, 5) expressed HEK293 cells. Compound 4 displayed affinity only α1β2γ2 subtype (Ki 31 nM), an in-depth, vivo study it revealed sedative anxiolytic-like properties without any amnesic myorelaxant effects rodents.

Abstract Background The mechanism underlying the pain symptoms associated with chemotherapeutic-induced peripheral neuropathy (CIPN) is poorly understood. Transient receptor potential ankyrin 1 (TRPA1), TRP vanilloid 4 (TRPV4), TRPV1, and oxidative stress have been implicated in several rodent models of CIPN-evoked allodynia. Thalidomide causes a painful CIPN patients via an unknown mechanism. Surprisingly, pathway responsible for such proalgesic response has not yet investigated animal...
