Wisteria floribunda agglutinin positive human Mac-2-binding protein (WFA(+)-hM2BP) was recently validated as a liver fibrosis glycobiomarker with fully automated lectin-antibody sandwich immunoassay. In this study, we supplied recombinant WFA(+)-hM2BP the standard glycoprotein and overlaid antibody to enhance robustness of quantification. The optimum conditions for producing were selected by cell glycome analysis based on lectin microarray. Interlot variability determined using an...
Despite the progress made in understanding glyco-alterations of specific glycoproteins such as α1-acid glycoprotein (AGP) associated with liver fibrosis, there has been no useful diagnostic assay a lectin recognizing fibrosis-specific alteration and an antibody against core protein. We therefore developed compatible multiple lectin-antibody sandwich immunoassay on basis results obtained by microarray analysis for monitoring fibrosis.AGP-enriched fractions derived from 0.5-μL sera 125...
Protein O-linked fucosylation is an unusual glycosylation associated with many important biological functions such as Notch signaling. Two pathways synthesizing O-fucosylglycans have been reported on cystein-knotted proteins, that is, epidermal growth factor-like (EGF-like) domains and thrombospondin Type 1 repeat (TSR) domains. We report here the molecular cloning characterization of a novel β1,3-glucosyltransferase (β3Glc-T) synthesizes Glcβ1,3Fucα- structure TSR domain. found...
Glycan alterations of proteins, a common feature cancer cells, are associated with carcinogenesis, invasion and metastasis. Glycomics, the study glycans glycan-binding proteins in various biological systems, is an emerging field postgenome postproteomics era. However, systematic robust strategies for glycomics still not fully established because structural analysis glycans, which comprise different patterns branching, possible linkage positions as well monomer anomericity, technically...

We previously proposed a high-throughput strategy to discover serological biomarker candidates of cancer. This focuses on series candidate glycoproteins that are specifically expressed in the original tissues (cells) target cancer and carry glycan structures associated with carcinogenesis [Narimatsu, H., et al. FEBS J.2010, 277(1), 95-105]. Here, we examined effectiveness our identifying biomarkers assess progression liver fibrosis for early detection hepatocellular carcinoma (HCC). On basis...
Epithelial ovarian cancer (EOC) is often asymptomatic and thus diagnosed at advanced stages with a poor prognosis. False-negative results for the conventional marker CA125 frequently occur in cases of clear cell carcinoma (CCC), type EOC; therefore, it necessary to develop biomarkers greater sensitivity. We previously reported strategy discover glycobiomarker candidates by combined lectin microarray IGOT–LC/MS analysis. have now optimized this discovering EOC biomarkers. Glycopeptides...
The importance of diagnosis and therapies for liver cirrhosis (LC) is indisputable. Thus, a reliable method monitoring the progression fibrosis resultant LC urgently needed. Previously, using lectin-assisted glycoproteomic method, we identified 26 serum glycoproteins as promising glycobiomarker candidates diseases. In this study, colony stimulating factor 1 receptor (CSF1R) marker candidate then established Wisteria floribunda agglutinin (WFA)-reactive CSF1R (WFA(+)-CSF1R) novel possible by...
Abstract The extent of liver fibrosis predicts prognosis and is important for determining treatment strategies chronic hepatitis. During the progression, serum levels Mac2 binding protein (M2BP) increase N-glycan structure changes to enable Wisteria floribunda agglutinin (WFA) lectin. As a novel diagnostic marker, glycosylation isomer M2BP (M2BPGi) has been developed. However, its glycan structures recognized by WFA are unclear. In this study, we analyzed site-specific using Glyco-RIDGE...
Serum hepatitis B surface antigen (HBsAg) is a component of both virus (HBV) virions and non-infectious subviral particles (SVPs). Recently, O-glycosylation the PreS2 domain middle HBsAg protein has been identified as distinct characteristic genotype C HBV versus SVPs. This study aimed to evaluate serum O-glycosylated levels in patients with chronic (CHB) treated nucleos(t)ide analogs (NAs).Forty-seven treatment-naïve CHB were retrospectively enrolled. at baseline after 48 weeks NA therapy...

Aims: The aim of this work was to develop a highly selective method detecting sulphate-reducing bacteria (SRB) in crude oil. Methods: A pair PCR primers designed based on an alignment the nucleotide sequence 16S rRNA genes from Desulfovibrionaceae family. DNA extraction oil performed by using zirconia beads and stool kit. Results: specifically detected Desulfovibrio Desulfomicrobium sediment sample. When nucleic acids extracted directly were used for PCR, 16SrRNAgenes Thermodesulforhabdus...
Wisteria floribunda agglutinin (WFA)-reactive ceruloplasmin (CP) is a candidate marker for ovarian clear cell carcinoma (CCC) reported in our previous paper. Herein, new measurement system was developed to investigate its potential as serum CCC. Site-specific glycome analysis using liquid chromatography/mass spectrometry showed that WFA-CP from CCC binds WFA via the GalNAcβ1,4GlcNAc (LDN) structure. We used mutant recombinant (rWFA), which has high specificity LDN structure, instead of...
Plasminogen activator inhibitor-1 (PAI-1) is a key regulator of the fibrinolytic system (1). The continuously high PAI-1 concentrations make fibrins resistant to dissolution by tissue-type plasminogen (tPA), which leads multiorgan dysfunction and bad prognosis in patients (2). Several methods have been developed for measuring functional activity plasma samples based on principle adding specified amount tPA excess residual after short incubation period (3). These methods, however, are neither...

A wheel-rail contact method for analyzing the train-structure nonlinear interaction that takes into account wheel and rail geometry is proposed in this article. The presented formulation divided three main parts: 1) geometric problem consisting of detection point calculation all variables needed determining forces; 2) normal which forces are determined based on Hertz theory; 3) tangential calculated using rolling friction laws by Kalker. Most existing methods treat directions as external...
