The site-directed chemical conjugation of antibodies remains an area great interest and active efforts within the antibody-drug conjugate (ADC) community. We previously reported a unique site modification using class immunoglobulin-G (IgG) Fc-affinity reagents to establish versatile, streamlined, site-selective native enhance therapeutic index resultant ADCs. This methodology, termed "AJICAP", successfully modified Lys248 produce site-specific ADC with wider than Food Drug...

A method for analyzing biothiols based on high-performance liquid chromatography (HPLC)-fluorescence detection under hydrophilic interaction (HILIC) conditions has been developed. Thiols were derivatized with nonfluorescent ammonium 7-fluoro-2,1,3-benzoxadiazole-4-sulfonate (SBD-F), which selectively reacts the thiol groups to furnish corresponding fluorescent SBD-thiols. Among six different kinds of HILIC columns examined, ZIC-HILIC column sulfobetaine in stationary phase proved be best...

Background: To delay the onset of dementia, it is important for healthy adults to take preventive actions before cognitive function clearly declines. Protein malnutrition a potential risk factor senile although precise link between protein/amino acid nutrition and unknown. The purpose this study was examine effect ingestion seven selected essential amino acids as granular powder, namely, leucine, phenylalanine, lysine supplemented with isoleucine, histidine, valine, tryptophan on...

Nutritional epidemiology shows that insufficient protein intake is related to senile dementia. The levels of in aged people are positively associated with memory function, and elderly high have a low risk mild cognitive impairment. Although the beneficial roles nutrition maintaining brain function well demonstrated, little known about mechanism by which dietary affects conditions. We fed mice diet (LPD) for 2 months, caused behavioral abnormalities, examined nutritional effect essential...

To realize efficient, fast separations on pillar array columns with turns, a novel turn low-dispersion and low-pressure-drop properties was developed. This "pillar-distribution-controlled" (PDC) designed as constant-radius filled octagonal pillars that were arranged to control the linear velocity of mobile phase in radial direction. After positions adjusted by computational fluid dynamics analysis, 27 mm long two turns fabricated 20 × mm2 silicon glass plate. The PDC suppressed sample...

High-performance liquid chromatography-fluorescence detection using a hydrophilic interaction chromatography-mode column (ZIC®-HILIC) was used to determine four kinds of thiol compounds in human serum. Sera were obtained from 34 subjects for this study (17 male aged 22-38 years and 17 female 18-38 years). Serum cysteine, cysteinylglycine, glutathione, γ-glutamylcysteine, derivatized with ammonium 7-fluoro-2,1,3-benzoxadiazole-4-sulfonate, separated on the ZIC®-HILIC quantified. The serum...

The chemical derivatization of target analytes can enhance the sensitivity and selectivity separation-based methods for metabolite analysis using microfluidic devices. However, development chromatography-based devices with integrated units is challenging. In this study, a novel unit pillar array (PA)-based mixing channel was developed postcolumn during on-chip liquid chromatography (LC). PA mixer enhanced between reagents in transverse direction, while preventing analyte dispersion flow...

Biological thiols – homocysteine (Hcy), cysteine (Cys), γ-glutamylcysteine (γGluCys), glutathione (GSH), and cysteinylglycine (CysGly) in plasma samples of cystathionine β-synthase (CBS, EC 4.2.1.22)-deficient mice were quantified using hydrophilic interaction liquid chromatography (HILIC) fluorescence detection. Mice deficient CBS provide a model mouse homocystinuria, an inherited metabolic disease characterized by the abnormal accumulation Hcy urine blood. For quantification, derivatized...

Hydrophilic interaction liquid chromatographic (HILIC) separation of biologically important thiols derivatized with 4-aminosulfonyl-7-fluoro-2,1,3-benzoxadiazole (ABD-F) was performed for the first time. After optimization mobile phase conditions, including acetonitrile content, pH, and buffer concentration, eight ABD-thiols (cysteamine, cysteine, N-acetylcysteine, glutathione, homocysteine, cysteinylglycine, γ-glutamylcysteine, N-(2-mercaptopropionyl)glycine as an internal standard) were...

In this study, we investigated suitable conditions for the reduction of disulfides in mouse serum samples by tris(2-carboxyethyl) phosphine (TCEP) fast analysis total biothiols. Disulfides were reduced with TCEP, and then, thiols derivatized fluorogenic reagent, ammonium 7-fluoro-2,1,3-benzoxadiazole-4-sulfonate (SBD-F). Interference peaks on chromatograms disappeared when TCEP reaction was conducted ice instead at room temperature, which is used classically. Low-molecular-weight disulfides,...

MEMS liquid chromatography (LC) microchip with low dispersion and a pressure drop turn structure was fabricated. In the structure, we used higher density pillar array at inner side while lower outer to control speed of sample flow. An LC chip an improved can obtain similar theoretical plate number (1233) compared previously proposed tapered (1135). The necessary inlet is also reduced from 2.7 0.4 MPa under flow rate 10 μL/min. These results show applicable for fast analysis time high conditions.
