Assembling for transcription initiation Eukaryotic by RNA polymerase II (Pol II) requires the assembly of a preinitiation complex (PIC) on core promoters. The binding TATA box–binding protein (TBP) to box promoter has been thought be general rule in PIC and initiation. However, most coding genes lack box, nearly all Pol II–mediated gene TBP-containing multisubunit factor IID (TFIID). Chen et al. determined structures human TFIID-based sequential states revealed that TFIID supports distinct...

Abstract DNMT1 is an important epigenetic regulator that plays a key role in the maintenance of DNA methylation. Here we determined crystal structure complex with USP7 at 2.9 Å resolution. The interaction between two proteins primarily mediated by acidic pocket and Lysine residues within DNMT1’s KG linker. This intermolecular required for USP7-mediated stabilization DNMT1. Acetylation linker impair DNMT1–USP7 promote degradation Treatment HDAC inhibitors results increase acetylated decreased...

UHRF1 is an important epigenetic regulator connecting DNA methylation and histone methylations. required for maintenance of through recruiting DNMT1 to replication forks. Recent studies have shown that the plant homeodomain (PHD) recognizes N terminus unmodified H3, interaction inhibited by H3R2, whereas tandem tudor domain (TTD) trimethylated H3 lysine 9 (H3K9me3). However, how two domains coordinately recognize methylations remains elusive. In this report, we identified PHD largely...

A maltose-functionalized metal–organic framework (MOF), MIL-101(Cr)-maltose, was developed via a simple two step post-synthetic modification of MIL-101(Cr)-NH2. With the use this nanomaterial, 33 glycopeptides were detected from digest human immunoglobulin G, demonstrating its high efficiency in glycoproteomic analysis. More importantly, generic functionalization route amino-derived MOFs opens new perspective material design sample preparation.

Pleiotropic transcription regulator RNA polymerase II (Pol II)–associated factor 1 (PAF1) governs multiple transcriptional steps and the deposition of several epigenetic marks. However, it remains unclear how ultimate outcome is determined by PAF1 whether relates to PAF1-controlled We use rapid degradation systems reveal direct functions in governing pausing partially recruiting Integrator-PP2A (INTAC), addition ensuring elongation. Following acute degradation, most destabilized undergoes...

Alzheimer's disease (AD), an age-associated neurodegenerative disorder, currently lacks effective clinical therapeutics. Traditional Chinese Medicine (TCM) holds promising potential in AD treatment, exemplified by Danggui Shaoyao San (DSS), a TCM formulation. The precise therapeutic mechanisms of DSS remain to be fully elucidated. This study aims uncover the efficacy and underlying AD, employing integrative approach encompassing gut microbiota metabolomic analyses.

Abstract The 5′-end capping of nascent pre-mRNA represents the initial step in RNA processing, with evidence demonstrating that guanosine addition and 2′-O-ribose methylation occur tandem early steps transcription by polymerase II, especially at pausing stage. Here, we determine cryo-EM structures paused elongation complex RNGTT, as well RNGTT CMTR1. Our findings show simultaneous presence NELF bound to II. exhibits two conformations, one which shows a notable rearrangement NELF-A/D compared...

Dynamin-related protein 1 (Drp1) is broadly expressed central nervous system plays a induction mitochondrial fission process. Spared nerve injury (SNI), due to dysfunction neurons within spinal dorsal horn (SDH), most common NP model. We explored neuroprotective Drp1 SDH SNI. Stachyose is a well-known gut microbiota-oriented prebiotic that could directly reach colon and influence the intestinal microenvironment. Through an in vitro fermentation on fecal microbiota from drug-naive new-onset perennial patient, we for first time revealed novel function of stachyose promoting microbial transformation tryptophan to indole catabolites (i.e., indole-3-propionic acid, indole-3-lactic acid indole-3-carboxaldehyde), which has shown improve functions glucose metabolism....
