The progressive deposition of misfolded hyperphosphorylated tau is a pathological hallmark tauopathies, including Alzheimer's disease. However, the underlying molecular mechanisms governing intercellular spreading species remain elusive. Here, we show that full-length soluble unconventionally secreted by direct translocation across plasma membrane. Increased secretion favored hyperphosphorylation, which provokes microtubule detachment and increases availability free protein inside cells....

Which isoforms of apolipoprotein E (apoE) we inherit determine our risk developing late-onset Alzheimer's Disease (AD), but the mechanism underlying this link is poorly understood. In particular, relevance direct interactions between apoE and amyloid-β (Aβ) remains controversial. Here, single-molecule imaging shows that all associate with Aβ in early stages aggregation then fall away as fibrillation happens. ApoE-Aβ co-aggregates account for ~50% mass diffusible aggregates detected frontal...

Transcription factors of the far‐upstream element‐binding protein (FBP) family represent cellular pathway hubs, and their overexpression in liver cancer (hepatocellular carcinoma [HCC]) stimulates tumor cell proliferation correlates with poor prognosis. Here we determine mode oncogenic FBP HCC cells. Using perturbation approaches (kinase inhibitors, small interfering RNAs) a novel system for rapalog‐dependent activation AKT isoforms, demonstrate that activity...

Tau is a soluble protein interacting with tubulin to stabilize microtubules. However, under pathological conditions, it becomes hyperphosphorylated and aggregates, process that can be induced by treating cells exogenously added tau fibrils. Here, we employ single-molecule localization microscopy resolve the aggregate species formed in early stages of seeded aggregation. We report entry sufficient assemblies into cytosol induces self-replication small doubling time 5 h inside HEK 1 day murine...

Background and study aims: Pancreatitis is the most common complication of therapeutic endoscopic retrograde cholangiopancreatography (ERCP), many pharmacoprophylactic approaches have been suggested, though not without controversy. The aim was to investigate impact combined therapy with diclofenac plus somatostatin on reducing frequency severity post-ERCP pancreatitis (PEP). Patients methods: A prospective, double-blind, placebo-controlled trial conducted in two tertiary...

Fibroblast growth factor 2 (FGF2) is a key signaling molecule in tumor-induced angiogenesis. FGF2 secreted by an unconventional secretory mechanism that involves phosphatidylinositol 4,5-bisphosphate-dependent insertion of oligomers into the plasma membrane. This process regulated Tec kinase-mediated tyrosine phosphorylation FGF2. Molecular interactions driving monomers membrane-inserted are unknown. Here we identify two surface cysteines critical for efficient secretion They represent...

FGF2 is exported from cells by an unconventional secretory mechanism. Here, we directly visualized individual membrane translocation events at the plasma using live cell TIRF microscopy. This process was dependent on both PI(4,5)P2–mediated recruitment of inner leaflet and heparan sulfates capturing outer leaflet. By simultaneous imaging appearance surface, revealed kinetics in living with average duration ∼200 ms. Furthermore, demonstrated oligomers a dimer being most prominent species. We...

Fibroblast growth factor 2 (FGF2) is a tumor cell survival that transported into the extracellular space by an unconventional secretory mechanism. Cell surface heparan sulfate proteoglycans are known to play essential role in this process. Unexpectedly, we found among diverse subclasses consisting of syndecans, perlecans, glypicans, and others, Glypican-1 (GPC1) principle rate-limiting drives secretion FGF2. By contrast, demonstrate GPC1 be dispensable for FGF2 signaling cells. We provide...

Here, we define structural elements are critical for physical interaction between and α1 subunit Na,K-ATPase. In intact cells, corresponding mutant forms were...

Super-resolution microscopy allows complex biological assemblies to be observed with remarkable resolution. However, the presence of uneven Gaussian-shaped illumination hinders its use in quantitative imaging or high-throughput assays. Methods developed circumvent this problem are often expensive, hard implement, not applicable total internal reflection fluorescence imaging. We herein demonstrate a cost-effective method overcome these challenges using small square-core multimodal optical...
