We present here a method for broadly characterizing single cells at the molecular level beyond more common morphological and transmitter/receptor classifications. The RNA from defined is amplified by microinjecting primer, nucleotides, enzyme into acutely dissociated region of rat brain. Further processing yields antisense RNA. A second round amplification results in greater than 10(6)-fold original starting material, which adequate analysis--e.g., use as probe, making cDNA libraries, etc....
Oxidative stress-induced granulosa cell (GCs) death represents a common reason for follicular atresia. Follicle-stimulating hormone (FSH) has been shown to prevent GCs from oxidative injury, although the underlying mechanism remains be elucidated. Here we first report that suppression of autophagic via some novel signaling effectors is engaged in FSH-mediated protection against damage. The decline viability caused by oxidant injury was remarkably reduced following FSH treatment, along with...
The melanoma cell line FO-1 does not express HLA class I antigens and acquire them on the surface after incubation with IFN-gamma. Immunochemical studies showed that cells synthesize heavy chain, but do beta 2-microglobulin (beta 2-mu). latter abnormality is associated lack of 2-mu mRNA which remains undetectable in incubated defect was identified as a genetic lesion B2m gene, since DNA hybridization analysis detected deletion first exon 5'-flanking region, segment intron gene. antigen...
Oxidative stress has been described as a prime driver of granulosa cell (GCs) death during follicular atresia. Increasing evidence suggests potential roles melatonin in protecting GCs from oxidative injury, though the underlying mechanisms remain largely undetermined. Here we first proposed that inhibition autophagy through some novel regulators contributes to melatonin-mediated survival under conditions stress. Oxidant-induced loss viability was significantly reduced after administration,...
It is widely accepted that inflammation may contribute to cognitive impairment in patients with vascular dementia (VD). Our prior clinical researches have reported acupuncture can alleviate function VD, but the underlying mechanisms are still unclear. The purpose of this research was explore whether alleviates by suppressing microRNA-93- (miR-93-) mediated Toll-like receptor (TLR) signaling pathway, which triggers inflammatory responses central nervous system. VD established permanent...
Synaptic transmission between light-sensory photoreceptor cells and downstream ON-bipolar neurons plays an important role for vertebrate vision. This process is mediated by the G-protein-coupled receptor pathway involving glutamate mGluR6 effector channel TRPM1. The signal occurs on a rapid timescale; however, molecular organization that ensures timely signaling in this cascade unknown. Genetic studies human patients animal models reveal cell depends synaptic protein nyctalopin. We have...
The time course of signaling via heterotrimeric G proteins is controlled through their activation by G-protein coupled receptors and deactivation the action GTPase accelerating (GAPs). Here we identify RGS7 RGS11 as key GAPs in mGluR6 pathway retinal rod ON bipolar cells that set sensitivity light-evoked responses. We showed using electroretinography single cell recordings elimination did not influence dark-adapted responses, but concurrent severely reduced magnitude dramatically slowed...
Type 2 diabetes and obesity are diseases related to surplus energy in the body. Abnormal interaction between hypothalamus adipose tissues is a key trigger of metabolism dysfunction. Extracellular vesicles (EVs) regulate intercellular communication by transporting intracellular cargo recipient cells thereby altering function cells. This study aimed evaluate whether adipocyte-derived EVs can act on hypothalamic neurons modulate intake identify EV-associated non-coding RNAs.Confocal imaging was...
Establishing synaptic contacts between neurons is paramount for nervous system function. This process involves transsynaptic interactions a host of cell adhesion molecules that act in cooperation with the proteins extracellular matrix to specify unique physiological properties individual connections. However, understanding molecular mechanisms generate functional diversity an input-specific fashion limited. In this study, we identify major components present cleft—members heparan sulfate...
Functional characterization of the GPCR interactome has been focused predominantly on intracellular interactions, yet GPCRs are increasingly found in complex with extracellular proteins. Extracellular leucine-rich repeat fibronectin type III domain containing 1 (ELFN1) was recently reported to physically anchor mGluR6 and mGluR7 across retinal hippocampal synapses, respectively; however, consequence transsynaptic interactions properties pharmacology these receptors unknown. In current study,...
Abstract Oxidative stress has been implicated in triggering granulosa cell (GC) death during follicular atresia. Recent studies suggested that follicle-stimulating hormone (FSH) a pivotal role protecting GCs from oxidative injury, although the exact mechanism remains largely unknown. Here, we report FSH promotes GC survival by inhibiting stress-induced mitophagy. The loss of viability caused was significantly reduced after treatment, which correlated with impaired activation mitophagy upon...
Members of the R7 family regulators G-protein signaling (R7 RGS) proteins form multi-subunit complexes that play crucial roles in processing light responses retinal neurons. The disruption these has been shown to lead loss temporal resolution photoreceptors and deficient synaptic transmission downstream Despite well established role one member this family, RGS9-1, controlling vertebrate phototransduction, organizational principles other members retina are poorly understood. Here we...
