Innate lymphoid cells (ILCs) can promote host defense, chronic inflammation, or tissue protection and are regulated by cytokines neuropeptides. However, their regulation diet microbiota-derived signals remains unclear. We show that an inulin fiber promotes Tph1-expressing inflammatory ILC2s (ILC2INFLAM) in the colon, which produce IL-5 but not tissue-protective amphiregulin (AREG), resulting accumulation of eosinophils. This exacerbates inflammation a murine model intestinal damage ILC2-...

ABSTRACT Microbial secondary metabolites, including isocyanide moieties, have been extensively mined for their repertoire of bioactive properties. Although the first naturally occurring (xanthocillin) was isolated from fungus Penicillium notatum over half a century ago, biosynthetic origins fungal isocyanides remain unknown. Here we report identification family synthases (ICSs) opportunistic human pathogen Aspergillus fumigatus . Comparative metabolomics overexpression or knockout mutants...

The maintenance of sufficient but nontoxic pools metal micronutrients is accomplished through diverse homeostasis mechanisms in fungi. Siderophores play a well established role for iron homeostasis; however, no copper-binding analogs have been found Here we demonstrate that, Aspergillus fumigatus, xanthocillin and other isocyanides derived from the xan biosynthetic gene cluster (BGC) bind copper, impact cellular copper content, significant metal-dependent antimicrobial properties....

Twenty-four metabolites, including seven new compounds (1–7), were isolated from the dried fruits of Psoralea corylifolia. On basis combined spectroscopic and chemical analysis, determined to be six flavonoids (1–6) a meroterpenoid (7). The absolute configurations natural products obtained, previously undetermined 16 17, assigned by several methods, such as NOE spectroscopy, optical rotation, CD spectroscopy. Several these exhibited moderate inhibitory activity toward Staphylococcus...

Five new manzamine alkaloids (1–5) and salt forms of two known manzamines (6 7), along with seven compounds (8–14) the same structural class, were isolated from an Indonesian Acanthostrongylophora sp. sponge. On basis results combined spectroscopic analyses, structure kepulauamine A (1) was determined to possess unprecedented pyrrolizine moiety, while others functional group variants manzamines. These exhibited weak cytotoxicity, moderate antibacterial activity, mild inhibition against...

Abstract Tumor heterogeneity is associated with the therapeutic failures of targeted therapies. To overcome such heterogeneity, a novel therapy proposed that could kill tumor populations diverse phenotypes by delivering nonselective cytotoxins to target‐positive cells as well surrounding via recurrent bystander killing effect. A representative prodrug prepared targets integrin αvβ3 and releases upon entering or caspase‐3. This allows αvβ3‐positive upregulate caspase‐3, which in turn,...

Abstract The genomes of many filamentous fungi, such as Aspergillus spp., include diverse biosynthetic gene clusters unknown function. We previously showed that low copper levels upregulate a cluster includes crmA , encoding putative isocyanide synthase. Here we show, using untargeted comparative metabolomics, CrmA generates valine-derived contributes to two distinct pathways under copper-limiting conditions.
