Abstract Cyclic guanosine monophosphate-adenosine monophosphate (cGAMP), produced by cyclic GMP-AMP synthase (cGAS), stimulates the production of type I interferons (IFN). Here we show that cGAMP activates DNA damage response (DDR) signaling independently its canonical IFN pathways. Loss cGAS dampens DDR induced genotoxic insults. Mechanistically, in a STING-TBK1-dependent manner, wherein TBK1 autophosphorylation kinase ATM, with consequent activation CHK2-p53-p21 signal transduction pathway...

Significance Alu elements, comprising more than 10% of the human genome, propagate via retrotransposition. This genomic expansion requires enzymatic activity L1 that reverse transcribes RNA into cDNA in nucleus. We report also undergoes L1-mediated transcription self-priming cytoplasm independent retrotransposition, providing evidence DNA synthesis this cellular compartment. newly discovered shunt molecule replication cycle induces death retinal pigmented epithelium, a hallmark atrophic...

The Prp43 DExD/H-box protein is required for progression of the biochemically distinct pre-messenger RNA and ribosomal (rRNA) maturation pathways. In Saccharomyces cerevisiae, Spp382/Ntr1, Sqs1/Pfa1, Pxr1/Gno1 proteins are implicated as cofactors necessary helicase activation during spliceosome dissociation (Spp382) rRNA processing (Sqs1 Pxr1). While otherwise dissimilar in primary sequence, these Prp43-binding each contain a short glycine-rich G-patch motif function thought to act or...

The level of insulin-like growth factor-binding protein 1 (IGFBP1), a liver-produced serum that regulates factor-I bioactivity, glucose homeostasis, and tissue regeneration, increases during inflammation. This manuscript describes novel pathway for the regulation hepatic IGFBP1 mRNA levels by interleukin (IL)-1β. Experiments with luciferase reporter system show IL-1β stimulates transcriptional activity from 1-kb promoter region IGFBP1. Although stimulation suppresses insulin activation...
