Acquiring neural signals at high spatial and temporal resolution directly from brain microcircuits decoding their activity to interpret commands and/or prior planning activity, such as motion of an arm or a leg, is prime goal modern neurotechnology. Its practical aims include assistive devices for subjects whose normal information pathways are not functioning due physical damage disease. On the fundamental side, researchers striving decipher code multiple which collectively make up nature's...

Introduction This study aimed to investigate the role of TAAR2-9 in sleep/wake regulation, given TAAR1's known involvement modulating neurotransmitter release and sleep patterns. Methods Male knockout (KO) wild-type (WT) mice were compared using baseline patterns, responses deprivation, effects TAAR1 agonists, dopaminergic markers. EEG recordings tyrosine hydroxylase immunohistochemistry used for analysis. Results KO exhibited lower delta theta power higher gamma power, with fragmented...

We have developed a prototype cortical neural sensing microsystem for brain implantable neuroengineering applications. Its key feature is that both the transmission of broadband, multichannel data and power required embedded microelectronics are provided by optical fiber access. The fiber-optic system aimed at enabling recording from rodents primates converting signals to digital stream infrared light pulses. In full whose performance summarized in this paper, an analog-to-digital converter...

Narcolepsy Type 1 (NT1), a sleep disorder with similar prevalence in both sexes, is thought to be due loss of the hypocretin/orexin (Hcrt) neurons. Several transgenic strains have been created model this and are increasingly being used for preclinical drug development basic science studies, yet most studies solely male mice. We compared narcoleptic symptomatology vs. female orexin-tTA; TetO-DTA mice, which Hcrt neuron degeneration can initiated by removal doxycycline (DOX) from diet. EEG,...

We discovered that optical stimulation of the mystacial pad in Emx1-Cre;Ai27D transgenic mice induces whisker movements due to activation ChR2 expressed muscles controlling retraction and protraction. Using high-speed videography anesthetized mice, we characterize amplitude protractions evoked by varying intensity, duration, frequency optogenetic stimulation. Recordings from primary somatosensory cortex (S1) indicated evokes robust yet longer latency responses than mechanical In head-fixed...

Summary Aversive symptoms, including insomnia experienced during opioid withdrawal, are a major drive to relapse; however, withdrawal‐associated sleep symptomatology has been little explored in preclinical models. We describe here model of withdrawal mice that resembles the phenotype characteristic humans. Male and female C57BL/6 were instrumented with telemeters record electroencephalogram, electromyogram, activity subcutaneous temperature. All received two treatments separated by 16‐day...

A multitude of neuroengineering challenges exist today in creating practical, chronic multichannel neural recording systems for primate research and human clinical application. Specifically, a) the persistent wired connections limit patient mobility from system, b) transfer high bandwidth signals to external (even distant) electronics normally forces premature data reduction, c) susceptibility infection due percutaneous nature implants all severely hinder success prosthetic systems. Here we...

Trace amine-associated receptor 1 (TAAR1) is a negative regulator of dopamine (DA) release. The partial TAAR1 agonist RO5263397 promotes wakefulness and suppresses NREM REM sleep in rodents non-human primates. We tested the hypothesis that TAAR1-mediated effects on sleep/wake regulation were due, part, to DA Male C57BL6/J mice (

Abstract Trace amine-associated receptor 1 (TAAR1) is known to negatively regulate dopamine (DA) release. Trace amine-associated receptor 1 (TAAR1) has gained attention for its roles in modulating neural systems, sleep/wake control, and as a therapeutic target neuropsychiatric disorders. Although TAARs 2-9 were initially identified non-canonical olfactory receptors, recent studies have extra-nasal distribution of multiple TAARs. To evaluate whether role arousal state regulation, we investigated control male TAAR2-9 knockout (KO) mice. After determination baseline patterns, the homeostatic...
