CRISPR/Cas9 genome editing is a very promising avenue for the treatment of variety genetic diseases. However, it still challenging to encapsulate machinery delivery. Protein N-myristoylation an irreversible co/post-translational modification that results in covalent attachment myristoyl-group N-terminus target protein. It serves as anchor protein associate with cell membrane and determines its intracellular trafficking activity. Extracellular vesicles (EVs) are secreted mediate cell-cell...

Genital mycoplasmas can break the cervical barrier and cause intraamniotic infection preterm birth. This study developed a six-chamber vagina-cervix-decidua-organ-on-a-chip (VCD-OOC) that recapitulates female reproductive tract during pregnancy with culture chambers populated by vaginal epithelial cells, stromal decidual cells. Cells cultured in VCD-OOC were characterized morphology immunostaining for cell-specific markers. We transferred media from cell chamber of to feto-maternal interface...

Adaptive human natural killer (NK) cells display significantly enhanced responsiveness to a broad-range of antibody-bound targets through the engagement CD16 compared conventional NK cells, yet direct reactivity against tumor is generally reduced. also distinct phenotype and differential expression numerous genes, including reduced signaling adapter FcRγ transcription factor PLZF. Human memory-like NK cells are commonly defined by either a lack of FcεRIγ or gain NKG2C expression. Here, we investigated the heterogeneity human CD56dim cell subpopulations according to expression and in large cohort (n=127). Although frequency FcεRIγ– NKG2C+ positively correlated, populations did not exactly overlap. The FcεRIγ+NKG2C+, FcεRIγ–NKG2C+, FcεRIγ–NKG2C–¬ were only evident after HCMV infection, but each had distinct characteristics. Among subpopulations, FcεRIγ–NKG2C+ exhibited...

Abstract “Adaptive” NK cells, characterized by FcRγ deficiency and enhanced responsiveness to Ab-bound, virus-infected have been found in certain hCMV-seropositive individuals. Because humans are exposed numerous microbes environmental agents, specific relationships between hCMV FcRγ-deficient cells (also known as g-NK cells) challenging define. Here, we show that a subgroup of rhesus CMV (RhCMV)-seropositive macaques possesses stably persist display phenotype resembling human cells....

Elderly people are at greater risk for complications from viral infections. A common explanation is that elderly have compromised immunity. Indeed, conservation of good natural killer (NK) cell cytotoxicity has been correlated with increased survival in later stages life. Our objective to examine how ageing affects NK homeostasis and their responses specific stimuli. We use flow cytometric methods the differences among cells adult (6 month) aged (22 month), male, C57/Bl6 mice. Herein, we...
