Ubiquitin chain complexity in cells is likely regulated by a diverse set of deubiquitinating enzymes (DUBs) with distinct ubiquitin preferences. Here we show that the polyglutamine disease protein, ataxin-3, binds and cleaves chains manner suggesting it functions as mixed linkage, chain-editing enzyme. Ataxin-3 through its amino-terminal Josephin domain carboxyl-terminal cluster interaction motifs neighboring pathogenic tract. both Lys48- or Lys63-linked yet preferentially Lys63 linkages....
Ubiquitin is a covalent signal that targets cellular proteins to the 26 S proteasome. Multiple ubiquitins can be ligated together through formation of isopeptide bonds between Lys48 and Gly76 successive ubiquitins. Such polyubiquitin chain constitutes highly effective proteolytic targeting signal, but its mode interaction with proteasome not well understood. Experiments address this issue have been limited by difficulties in preparing useful quantities chains uniform...
SAGA is a transcriptional coactivator complex that conserved across eukaryotes and performs multiple functions during activation elongation. One role deubiquitination of histone H2B, this activity resides in distinct subcomplex called the deubiquitinating module (DUBm), which contains ubiquitin-specific protease Ubp8, bound to Sgf11, Sus1, Sgf73. The depends on presence all four DUBm proteins. We report here 1.90 angstrom resolution crystal structure ubiquitin aldehyde, as well 2.45...

In at least nine inherited diseases polyglutamine expansions cause neurodegeneration associated with protein misfolding and the formation of ubiquitin-conjugated aggregates. Although expanded triggers disease, functional properties host proteins also must influence pathogenesis. Using complementary in vitro cell-based approaches we establish that disease protein, ataxin-3, is a poly-ubiquitin-binding protein. stably transfected neural cell lines, normal ataxin-3 both co-precipitate...
Invertase, extracted from broken cells of Saccharomyces cerevisiae X-2180 mm2 mannan mutant, was separated into a fraction insoluble in 75% ammonium sulfate (P75 invertase, 36% carbohydrate) and soluble (S75 53% carbohydrate). The latter reacted with antibodies specific for the alpha 1 leads to 6-linked mannose mannoprotein outer chain, whereas P75 invertase failed react this antiserum although it did serum against terminal 3-linked units that are characteristic core. A bacterial endo...
The specificity of the ubiquitin (Ub) isopeptidase in PA700 regulatory complex bovine 26 S proteasome was investigated. Disassembly poly-Ub by this enzyme is restricted to distal-end Ub substrate, i.e. farthest from site protein attachment poly-Ub-protein conjugates. determinants recognized were probed use mutant ubiquitins incorporated into Lys48-linked substrates. could not disassemble chains that contained a distal Ub(L8A,I44A). This suggested either interacts directly with Leu8 or Ile44...
