EZH2 is overexpressed in poor-prognostic chronic lymphocytic leukaemia (CLL) cases, acting as an oncogene; however, thus far, the target genes CLL have not been disclosed. In this study, using ChIP-sequencing, we identified and H3K27me3 two prognostic subgroups of with distinct prognosis outcome, i.e., cases unmutated (U-CLL, n = 6) or mutated IGHV (M-CLL, 6). While majority oncogenic pathways were equally enriched for both subgroups, PI3K pathway differentially bound by U-CLL versus M-CLL....
Certain subtypes of acute myeloid leukemia (AML) in children have inferior outcome, such as AML with translocation t(7;12)(q36;p13) leading to an MNX1::ETV6 fusion along high expression MNX1. We identified the transforming event this and possible ways treatment. Retroviral MNX1 was able induce mice, similar gene pathway enrichment t(7;12) patient data. Importantly, only induced immune incompetent mice using fetal but not adult hematopoietic stem progenitor cells. The restriction capacity...
Abstract Sterile alpha motif and histidine-aspartate (HD) domain–containing protein 1 (SAMHD1) is a deoxynucleoside triphosphate triphosphohydrolase with ara-CTPase activity that confers cytarabine (ara-C) resistance in several hematological malignancies. Targeting SAMHD1’s has recently been demonstrated to enhance ara-C efficacy acute myeloid leukemia. Here, we identify the transcription factor SRY-related HMG-box containing 11 (SOX11) as novel direct binding partner first known endogenous...
To survive chemotherapy, lymphoma cells can relocate to protective niches where they receive support from the non-malignant cells. The biolipid 2-arachidonoylglycerol (2-AG), an agonist for cannabinoid receptors CB1 and CB2, is released by stromal in bone marrow. investigate role of 2-AG lymphoma, we analyzed chemotactic response primary B-cell enriched peripheral blood twenty-two chronic lymphocytic leukemia (CLL) five mantle cell (MCL) patients towards alone and/or chemokine CXCL12....
The Ten-eleven-translocation 1 (TET1) protein is a member of dioxygenase family that catalyzes the oxidation 5-methylcytosine to 5-hydroxymethylcytosine. TET1 differentially expressed in many cancers, including leukemia. However, very little known about mechanism behind deregulation. Previously, by characterizing global methylation patterns CLL patients using MBD-seq, we found as one methylated regions with gene-body hypermethylation. Herein, characterize mechanisms control gene activity at...
The sterile alpha motif and histidine-aspartate (HD) domain containing protein 1 (SAMHD1) is a deoxynucleoside triphosphate triphosphohydrolase with ara-CTPase activity that confers cytarabine (ara-C) resistance in several haematological malignancies. Targeting SAMHD1's has recently been demonstrated to enhance ara-C efficacy acute myeloid leukemia. Here, we identify the transcription factor SRY-related HMG-box 11 (SOX11) as novel direct binding partner first known endogenous inhibitor of...
Abstract SAMHD1 is a deoxynucleoside triphosphate triphosphohydrolase (dNTPase) that restricts viral replication in infected cells and limits the sensitivity to cytarabine by hydrolysing its active metabolite, as recently shown acute myeloid leukemia. Cytarabine an essential component Nordic mantle cell lymphoma protocols (MCL2 MCL3) for induction high-dose chemotherapy treatment before autologous stem transplantation younger patients with (MCL). We here investigated expression of...

Abstract Certain subtypes of acute myeloid leukemia (AML) in children have still inferior outcome. One these AML has a translocation t(7;12)(q36;p13), always leading to high expression MNX1 and often :: ETV6 fusion expression. Here we identified the transforming event this possible ways target them. Only was able induce mice, observed using hematopoietic stem progenitor cells derived from fetal origin but not adult bone marrow. The restriction capacity liver is concordance with fact that...
