Abstract Background Aggregation of α-synuclein (α-syn) is a prominent feature Parkinson’s disease (PD) and other synucleinopathies. Currently, α-syn seed amplification assays (SAAs) using cerebrospinal fluid (CSF) represent the most promising diagnostic tools for However, CSF itself contains several compounds that can modulate aggregation in patient-dependent manner, potentially undermining unoptimized SAAs preventing quantification. Methods In this study, we characterized inhibitory effect...

Recent structural studies show distinct morphologies for the fibrils of Aβ(1-42) and Aβ(1-40), which are believed not to co-fibrillize. We describe here a novel, structurally-uniform 1 : mixed fibrillar species, differs from both pure fibrils. It forms preferentially even when Aβ(1-40) peptides in non-stoichiometric ratio.

Background: The prion-like misfolding and aggregation of α-synuclein (α-syn) is involved in the pathophysiology Parkinson's disease other synucleinopathies. Seed amplification assays (SAAs) are biophysical tools that take advantage on peculiar properties prion proteins by amplifying small amounts aggregates biological fluids at expense recombinant monomeric protein added solution. SAAs have emerged as most promising for diagnosis synucleinopathies vivo. However, diagnostic outcome depends...

Today it is widely recognized that the PD-1/PD-L1 axis plays a fundamental role in escaping immune system cancers, so anti-PD-1/PD-L1 antibodies have been evaluated for their antitumor properties more than 1000 clinical trials. As result, some of them entered market revolutionizing treatment landscape specific cancer types. Nonetheless, new era based on development small molecules as anti PD-L1 drugs has begun. There are, however, limitations to advancing these compounds into stages...

Biocompatible hydrogels for tissue regeneration/replacement and drug release with specific architectures can be obtained by three-dimensional bioprinting techniques. The preservation of the higher order structure proteins embedded in as drugs or modulators is critical their biological activity.
