The interplay between cancer cells and immune is a key determinant of tumor survival. Here, we uncovered how tumors exploit the immunomodulatory properties extracellular matrix to create microenvironment that enables their escape from surveillance. Using orthotopic grafting mammary in immunocompetent mice autochthonous models breast cancer, discovered tenascin-C, molecule absent most healthy adult tissues but expressed at high levels associated with poor patient prognosis many solid cancers,...
Glycosaminoglycans (GAGs) are key ligands for proteins involved in physiological and pathological processes. Specific GAG-binding patterns rarely identified, with the heparin pentasaccharide as an Antithrombin-III ligand being best characterized. Generating glycan-specific antibodies is difficult due to their size, pattern dispersion, flexibility. Single-domain variable new antigen receptors (VNAR nanobodies) from nurse sharks highly soluble, stable, versatile. Their unique properties...
Chemokine binding to glycosaminoglycans (GAGs) is recognised be an important step in inflammation and other pathological disorders like tumor growth metastasis. Although different ways strategies interfere with these interactions are being pursued, no major breakthrough the development of glycan-targeting drugs has been reported so far. We have engineered CXCL8 towards a dominant-negative form this chemokine (dnCXCL8) which was shown highly active various inflammatory animal models due its...
We have engineered GPCR (G protein-coupled receptor) knock-out and high GAG-binding affinity into CXCL12α to inhibit CXCL12α-induced cell migration. Compared wtCXCL12, the mutant (Δ8 L29K V39K) exhibited a 5.6-fold 2.2-fold increase for heparin heparan sulfate, respectively. From NaCl-based displacement chromatography we concluded that more amino acid replacements would lead altered GAG (glycosaminoglycan) ligand specificity. silencing by this was shown in murine seeding model of human...
Bidirectional dialogue between cellular and non-cellular components of the tumor microenvironment (TME) drives cancer survival. In extracellular space, combinations matrix molecules soluble mediators provide external cues that dictate behavior TME resident cells. Often studied in isolation, integrated from complex tissue microenvironments likely function more cohesively. Here, we study interplay molecule tenascin-C (TNC) chemokine CCL2, both elevated associated with progression breast...
Tenascin-C (TNC) is a complex glycoprotein of the extracellular matrix (ECM) involved in plethora (patho-)physiological processes, such as oncogenesis and inflammation. Since chemokines play an essential role both disease we have investigated here binding TNC to some key chemokines, namely CCL2, CCL26, CXCL8, CXCL10, CXCL12. Thereby, differential chemokine-TNC pattern was observed, with CCL26 exhibiting highest CCL2 lowest affinity for TNC. Heparan sulfate (HS), another member ECM, proved be...
ABSTRACT Members of the MOAG/SERF protein family have been attributed a neuropathologic significance because their ability to enhance proteotoxic polymerization amyloidogenic proteins such as alpha-Synuclein (aSyn). However, cellular function remains unknown. Here, we identify SERF1a an RNA-organizing i) with no structural homology canonical RNA-binding proteins, ii) RNA-chaperoning activity which favours incorporation RNA into nucleoli and liquid-like artificial RNA-organelles, iii) high...
Abstract The chemokine CXCL10 is released by a plethora of cells, including immune and metastatic cancer following stimulation with interferon-gamma. It acts via its GPC receptor on T-cells attracting them to various target tissues. Glycosaminoglycans (GAGs) are regarded as co-receptors chemokines, which enable the establishment chemotactic gradient for cell migration. We have engineered human towards improved T-cell mobilisation implementing single site-directed mutation N20K into protein,...
