Background Human plasma and serum are widely used matrices in clinical biological studies. However, different collecting procedures the coagulation cascade influence concentrations of both proteins metabolites these matrices. The effects on metabolite concentration profiles have not been fully characterized. Methodology/Principal Findings We analyzed 163 samples collected simultaneously from 377 fasting individuals. To ensure data quality, 41 with low measurement stability were excluded...

Side-chain modeling has a widespread application in many current methods for protein tertiary structure determination, prediction, and design. Of the existing side-chain methods, rotamer-based are fastest most efficient. Classically, rotamer is conceived as single, rigid conformation of an amino acid side-chain. Here, we present flexible model which continuous ensemble conformations that cluster around classic rotamer. We have developed thermodynamically based method calculating effective...

A seven‐iron ferredoxin was isolated from aerobically grown cells of the hyperthermoacidophilic archaeon Desulfurolobus ambivalens (DSM 3772). The protein is monomeric, with an apparent molecular mass 15 kDa and contains 7 iron atoms/molecule. N‐terminal sequence shows a large similarity (70% identity) that Sulfolobus acidocaldarius. EPR characteristics in both native (oxidized) dithionite‐reduced states this allowed unequivocal identification [3Fe‐4S] 1+/0 center, reduction potential −270 ±...

With the objective of improving side-chain conformation prediction, we have analyzed influence various factors on prediction by Self-Consistent Mean Field Theory method, applied to a set high resolution x-ray protein structure models. These may be classed as variations in mean field optimization protocol, potential energy function, and rotamer library completeness. We developed an protocol that consistently reached lower conformational free energies than two other protocols. This led...

Sub-nanosecond bipolar high voltage pulses are a very important tool for food processing, medical treatment, waste water and exhaust gas processing. A Hybrid Modulator sub-microsecond pulse generation, comprising an unipolar solidstate Marx generator connected to load through stack Blumlein system that produces further multiplies the amplitude, is presented. Experimental results from assembled prototype show generation of 1000 V amplitude with 100 ns width 1 kHz repetition rate.
