Direct, rapid, sensitive, and selective detection of nucleic acids in complex biological fluids is crucial for medical early diagnosis. We herein combine the trans-cleavage ability clustered regularly interspaced short palindromic repeats (CRISPR)/Cas12a with Au-nanobeacon to establish a CRISPR-based biosensor, providing rapid miRNA high speed attomolar sensitivity. In this strategy, we first report that activity CRISPR/cas12a, which was previously reported be triggered only by target ssDNA...

The current state of cancer therapy encourages researchers to develop novel efficient nanocarriers. Halloysite nanotubes (HNTs) are good nanocarrier candidates due their unique nanoscale (40-80 nm in diamter and 200-500 length) hollow lumen, as well biocompatibility low cost. In our study, we prepared a type folate-mediated targeting redox-triggered anticancer drug delivery system, so that Doxorubicin (DOX) can be specifically transported tumor sites the over-expressed folate-receptors on...

The precise detection of extracellular ATP, although a challenging task, is great significance for understanding the related cell processes. Herein, we developed ratiometric DNA nanoswitch by employing tweezer and split aptamer. composed three specially designed ssDNA strands, namely, central strands O1, O2, O3. This can be anchored on membrane cholesterol labeled at 3' end Initially, adopts an open state, separating dual fluorophores giving rise to low FRET (fluorescence resonance energy...

We, herein, indicated for the first time Rnase activities of LbCas12a on linear ssRNA above 11 bases, and hairpin RNA substrates. Meanwhile, bound to ssDNA or exhibited different cleavage efficiencies various substrates, including short ssDNA, DNA, RNA. With DNA as a reporter, we attained detection limit 5 pM 50 targets, respectively. We believe that these findings will pave new avenue expanding reporter toolbox Cas12a-based diagnostics in biosensing biochemistry.

In this work, we present a highly sensitive, specific, and versatile method to quantify miRNA expression by coupling CRISPR-Cas12a with cyclic reverse transcription (CRT), termed as CRISPR-CRT. Each target was first converted amplified into multiple hairpin RT products via CRT. Afterward, the could serve activators initiate collateral cleavage activity of CRISPR-Cas12a. Due above two-stage amplification, assay detect at sub-femtomolar level (LOD, 0.201 fM). Since sequence is double checked:...

A water-compatible molecularly imprinted polymer was prepared by Pickering emulsion polymerization using halloysite nanotubes as stabilized solid particles. During polymerization, we used 4-vinylpyridine monomer, divinylbenzene cross-linking agent, toluene porogen, 2,2-azobisisobutyronitrile initiator, 2,4-dichlorophenoxyacetic acid template to form the oil phase, and Triton X-100 aqueous solution water phase. The characterized Fourier transform infrared spectroscopy scanning electron...
