Abstract Mutations in Cullin-3 ( Cul3 ), a conserved gene encoding ubiquitin ligase, are strongly associated with autism spectrum disorder (ASD). Here, we characterize ASD-related pathologies caused by neuron-specific knockdown Drosophila . We confirmed that neuronal causes short sleep, paralleling sleep disturbances ASD. Because defects and ASD linked to metabolic dysregulation, tested the starvation response of flies; they starved faster had lower triacylglyceride levels than controls,...
Longitudinal measurements of food intake remain a challenge in Drosophila studies nutrition and behavior. Here, we report an improved method for measuring fly using dye-labeled excreta quantification (EX-Q). Reducing the surface area medium maximized recovery accuracy estimating total consumption. The EX-Q is compatible with agar-based makes it possible to measure consumption over extended period at multiple time points without sacrificing flies. Using EX-Q, revealed nutrient- age-specific...
Taste drives appropriate food preference and intake. In Drosophila, taste neurons are housed in both external internal organs, but the latter have been relatively underexplored. Here, we report that Poxn mutants with a minimal system of pharyngeal can avoid many aversive tastants, including bitter compounds, acid, salt, suggesting is sufficient for rejecting intake compounds. Optogenetic activation selected during feeding events elicits changes parameters suppress Functional dissection...
Background and aim Social factors may be of importance causally act as maintenance in patients with anorexia nervosa. Oxytocin is a neuromodulatory hormone involved social emotional processing associated attentional processes. This study aimed to examine the impact oxytocin on processes faces representing anger, disgust, happiness Method A double-blind, placebo-controlled within-subject crossover design was used. Intranasal or placebo followed by visual probe detection task depicting...
Perceived palatability of food controls caloric intake. Sweet taste is the primary means detecting carbohydrate content food. Surprisingly, sweet sensitivity responsive to extrinsic factors like diet, and this occurs by unknown mechanisms. Here, we describe an unbiased proteomic investigation into in fruit fly. We identify a dopamine/cyclic AMP (cAMP)/CREB axis acting within neurons that perception but largely dispensable for acute transduction. This pathway modulates response both sensory-...
This study aimed to evaluate the antioxidant activities of Tenebrio molitor L. (TM) and Protaetia brevitarsis seulensis (PB). Water extracts TM PB were prepared at concentrations 2, 4, 6, 8, 10, 20, 40, 60, 80, 100 mg/mL. Cell viability assays performed cytoprotective effects water extracts. Antioxidant measured using ABTS DPPH radical scavenging reducing power. The cell significantly increased in a dose-dependent manner, with exhibiting superior efficacy compared PB. exhibited high...
Abstract Neurofibromatosis type 1 is a chronic multisystemic genetic disorder that results from loss of function in the neurofibromin protein. Neurofibromin may regulate metabolism, though underlying mechanisms remain largely unknown. Here we show regulates metabolic homeostasis Drosophila via discrete neuronal circuit. Loss increases rate Ras GAP-related domain-dependent mechanism, feeding homeostatically, and alters lipid stores turnover kinetics. The increase independent locomotor...
We describe the theoretical rationale and protocol for Self-Help And Recovery guide Eating Disorders (SHARED), a trial investigating whether guided self-care intervention (Recovery MANTRA) is useful addition to treatment as usual individuals with anorexia nervosa. MANTRA, 6-week supplemented by peer mentorship, module extension of Maudsley Model Treatment Adults Anorexia Nervosa targets maintenance factors identified cognitive-interpersonal model illness. Patients accessing outpatient...
Plant-derived caffeine is regarded as a defensive compound produced to prevent herbivory. Caffeine generally repellent insects and often used study the neurological basis for aversive responses in model insect, Drosophila melanogaster. also studied its stimulatory properties where sleep or drowsiness suppressed across range of species. Since limiting access food inhibits fly sleep—an effect known starvation-induced suppression—we tested whether aversion caffeinated results reduced nutrient...
Sexually experienced female rats return to the male more quickly after intromissions, exhibit shorter interintromission intervals, and spend time with rat during a test of paced mating behavior in comparison naïve rats. The present study tested whether these changes reflect heightened sexual motivation independent receipt vaginocervical stimulation and/or neurochemical medial preoptic area (mPOA). Ovariectomized, were given estradiol benzoate progesterone, then received either 6 encounters...
Factors that mediate ethanol preference in Drosophila melanogaster are not well understood. A major confound has been the use of diverse methods to estimate consumption. We measured fly consumptive on base diets varying nutrients, taste and concentration. Both sexes showed an was abolished high nutrient concentration diets. Additionally, manipulating total food intake without altering nutritive value diet or sufficient evoke eliminate preference. Absolute volume consumed were stronger...
The prototypical M13 peptidase, human Neprilysin, functions as a transmembrane "ectoenzyme" that cleaves neuropeptides regulate e.g. glucose metabolism, and has been linked to type 2 diabetes. family undergone remarkable, conserved, expansion in the Drosophila genus. Here, we describe function of melanogaster Neprilysin-like 15 (Nepl15). Nepl15 is likely be secreted protein, rather than protein. changes critical catalytic residues are conserved across genus renders protein catalytically...
Sleep and feeding are vital homeostatic behaviors, disruptions in either can result substantial metabolic consequences. Distinct neuronal manipulations
In previous work, we found that short sleep caused sensitivity to oxidative stress; here set out characterize the physiological state of a diverse group chronically short-sleeping mutants during hyperoxia as an acute stress. Using RNA-sequencing analysis, had normal transcriptional stress response relative controls. both and controls, led downregulation glycolytic genes upregulation involved in fatty acid metabolism, reminiscent metabolic shifts sleep. We hypothesized may be sensitive...
ABSTRACT Neurofibromatosis type 1 (NF1) is a genetic disorder predisposing patients to range of features, the most characteristic which include areas abnormal skin pigmentation and benign tumors associated with peripheral nerves, termed neurofibromas. Less common, but more serious symptoms also malignant nerve sheath tumors, other malignancies, learning disabilities. The NF1 gene encodes neurofibromin, large protein that functions as negative regulator Ras signaling mediates pleiotropic...
