Fucose, the monosaccharide frequent in N- and O-glycans, is a part of Lewis-type antigens that are known to mediate direct sperm binding zona pellucida. Such interaction was found be inhibited vitroby fucose-containing oligo- polysaccharides, as well neoglycoproteins. The objective this study screen seminal plasma proteins infertile/subfertile men for content density fucosylated glycoepitopes, compare them samples fertile normozoospermic subjects. Seminal were separated polyacrylamide gel...

The expression and activity of matrix metalloproteinases (MMPs) may be regulated by oxidative stress in various pathophysiological processes; therefore, the aim present study was to analyse associations between gelatinases MMP-9 MMP-2 their tissue inhibitors TIMP-1, TIMP-2 levels total antioxidant capacity (TAC) advanced oxidation protein products (AOPP) seminal plasma prepared for artificial insemination. Levels MMPs TIMPs were evaluated using ELISA, whereas TAC AOPP 131 childless men 38...

The impact of seminal plasma components on the fertilization outcomes in humans is still under question. increasing number couples facing problems with conception raises need for predictive biomarkers. Detailed understanding molecular mechanisms accompanying remains another challenge. Carbohydrate-protein recognition may be key importance this complex field. In study, we analyzed unique glycosylation pattern proteins, display high-mannose and hybrid-type oligosaccharides, by means their...

According to a concept of fetoembryonic defence, protein-carbohydrate interaction may be involved in the regulation maternal immunity that prevents rejection allograft spermatozoa, embryo and fetus. In present study we focussed on evaluation expression glycoepitopes crucial importance this process: LewisY (LeY) LewisX (LeX) as well terminal sialylation. Polyacrylamide gel electrophoresis with sodium dodecyl sulphate was used separate seminal plasma samples fertile (n=10) infertile (n=103)...

Increased male age is associated with a significant reduction in semen quality. Little known about the sperm proteome changes resulting from aging process. This study aimed to investigate relationship between functional quality and of epididymal spermatozoa dogs that were differing age. The was conducted on 30 divided into three groups. G1-12 41 months old, G2-42 77 G3-78 132 old. samples assessed using computer-assisted analysis (CASA). proteins analyzed gel electrophoresis (SDS-PAGE),...

We report application of lectin microarrays, exploiting simultaneous interaction seminal plasma samples with multiple lectins different sugar specificities to compare the glycomes fertile and infertile men. The results indicate reduced reactivity associated decreased fertility, especially affecting oligozoospermic subjects probably O-glycosylation. Lectin microarrays may become a potent tool for semen analysis in search association glycosylation male fertility.

This study was provided for proteomic analysis of intracellular and membrane-associated fractions canine (Canis lupus familiaris) epididymal spermatozoa additionally to find optimal sonication parameters the sperm morphological structure separation protein isolation. Sperm samples were collected from 15 dogs. fractions: (SIPs) (SMAPs) isolated. After sonication, morphology evaluated using Spermac Stain™. The analyzed gel electrophoresis (SDS-PAGE) nanoliquid chromatography coupled quadrupole...

Sperm maturation in the epididymis is based on interactions with proteins from epididymal fluid (EF). The aim of study was to profile canine EF proteome and investigate correlations between protein content spermatozoa (ES) motion parameters. Twenty-three male dogs were divided into two groups: good sperm motility (GSM) poor (PSM). total progressive differed significantly (p = 0.031; p < 0.001, respectively) GSM group PSM group. semen samples centrifuged separate apart ES. analyzed using...
