Abstract The CRISPR–Cas9 system provides a versatile toolkit for genome engineering that can introduce various DNA lesions at specific genomic locations. However, better understanding of the nature these and repair pathways engaged is critical to realizing full potential this technology. Here we characterize different arising from each Cas9 variant resulting pathway engagement. We demonstrate presence polarity overhang structure determinant double-strand break choice. Similarly, single nicks...

Adults often develop chronic immune thrombocytopenia (ITP) for which treatment order is uncertain. Rituximab and three cycles of dexamethasone (4R + 3Dex) improve responses short-term disease control but long-term outcome not known. In adults with ITP treated 4R 3D, we sought associated prognostic variables. Forty-nine at Weill-Cornell received 3Dex. Their clinical characteristics were reviewed. Duration was median time to failure; Kaplan-Meier estimates developed. Vbeta Tcell receptor...

Brief motivational interventions (BMIs) are usually effective for reducing alcohol use and consequences in primary care settings. We examined readiness to change drinking as a mediator of the effects BMI on alcohol-related consequences. Participants were randomized into three conditions: (a) standard plus assessment (SC), (b) SC (BI), (c) BI booster session (BIB). At 12-month follow-up BIB patients had significantly reduced more than patients. Patients receiving or maintained higher scores 3...

This study investigates the relationship between level of acculturation and stress extent to which each predicts problems related drinking. Hispanics who met criteria for hazardous drinking completed measures acculturation, stress, problems. Sequential multiple regression was used determine whether levels self-reported predicted concurrent alcohol after controlling predictive value level. Acculturation accounted a significant variance in problems, while adjusting income, education. Choosing...
