Oxidative DNA damage is recognized by 8-oxoguanine (8-oxoG) glycosylase 1 (OGG1), which excises 8-oxoG, leaving a substrate for apurinic endonuclease (APE1) and initiating repair. Here, we describe small molecule (TH10785) that interacts with the phenylalanine-319 glycine-42 amino acids of OGG1, increases enzyme activity 10-fold, generates previously undescribed β,δ-lyase enzymatic function. TH10785 controls catalytic mediated nitrogen base within its molecular structure. In cells, OGG1...

Endocrine-disrupting chemicals (EDCs) generate reproductive dysfunctions affecting the biosynthesis of steroid hormones and genes steroidogenic pathway. EDCs effects are mainly reported as a result exposure to single compounds. However, humans environmentally exposed mixture EDCs. Herein, we assess chronic alkylphenols phthalates versus in mouse testes histology steroidogenesis. Pregnant mice were through drinking water to: 0.3 mg/kg-body weight (BW)/d each phthalate (bis (2-ethylhexyl)...

Classical non-homologous end joining (NHEJ) is a molecular pathway that detects, processes, and ligates DNA double-strand breaks (DSBs) throughout the cell cycle. Mutations in several NHEJ genes result neurological abnormalities immunodeficiency both humans mice. The required for V(D)J recombination developing B T lymphocytes, class switch mature cells. Ku heterodimer formed by Ku70 Ku80 recognizes DSBs facilitates recruitment of accessory factors (e.g., DNA-PKcs, Artemis, Paxx Mri/Cyren)...

DNA double-strand breaks (DSBs) trigger the Ataxia telangiectasia mutated (ATM)-dependent damage response (DDR), which consists of histone H2AX, MDC1, RNF168, 53BP1, PTIP, RIF1, Rev7, and Shieldin. Early stages B T lymphocyte development are dependent on recombination activating gene (RAG)-induced DSBs that form basis for further V(D)J recombination. Non-homologous end joining (NHEJ) pathway factors recognize, process, ligate DSBs. Based numerous loss-of-function studies, DDR were thought to...

Spermatogonial stem cell (SSC) is known for its self-renewal capacity. We have studied the in vitro proliferation of isolated SSC from adult alpaca (Vicugna pacos) testes. A total 107 samples were evaluated which 31 at baseline, 36 cultivated DMEM and 40 STEMPRO media. Half was analysed after 14 days, rest 21 days. Round subpopulations identified with FITC-DBA by flow cytometry: strongly positive DBA (sDBA+) as SSC, weakly (wDBA+) early differentiation negative differentiated. At beginning,...
