Ascorbic acid (AsA) maintains redox homeostasis by scavenging reactive oxygen species from prokaryotes to eukaryotes, especially plants. The enzyme monodehydroascorbate reductase (MDHAR) regenerates AsA catalysing the reduction of monodehydroascorbate, using NADH or NADPH as an electron donor. detailed recycling mechanism MDHAR remains unclear due lack structural information. Here, we present crystal structures in presence cofactors, nicotinamide adenine dinucleotide (NAD+) and phosphate...

Abstract Dehydroascorbate reductase (DHAR) is a key enzyme involved in the recycling of ascorbate, which catalyses glutathione (GSH)-dependent reduction oxidized ascorbate (dehydroascorbate, DHA). As result, DHAR regenerates pool reduced and detoxifies reactive oxygen species (ROS). In previous experiments involving transgenic rice, we observed that overexpression enhanced grain yield biomass. Since structure not available, enzymatic mechanism well-understood remains poorly characterized. To...

A novel microbial esterase, EaEST, from a psychrophilic bacterium Exiguobacterium antarcticum B7, was identified and characterized. To our knowledge, this is the first report describing structural analysis biochemical characterization of an esterase isolated genus Exiguobacterium. Crystal structure determined at resolution 1.9 Å, showed that enzyme has canonical α/β hydrolase fold with α-helical cap domain catalytic triad consisting Ser96, Asp220, His248. Interestingly, active site EaEST...

A hyperthermophilic membrane-related β-1,4-endoglucanase (family 5, cellulase) of the archaeon Pyrococcus horikoshii was found to be capable hydrolysing cellulose at high temperatures. The cellulase has promise for applications in biomass utilization. To clarify its detailed function, we determined crystal structures mutants enzyme complex with either substrate or product ligands. We were able resolve different kinds 1.65–2.01 Å (1 Å=0.1 nm). structural analysis various mutant enzymes...

Glutamate decarboxylase (GAD) from the archaeon Pyrococcus horikoshii was successfully expressed and purified, with aim of developing a hyperthermostable GAD for industrial applications. Its biochemical properties were different those reported other GADs. The enzyme had broad substrate specificity, its optimum pH temperature 8.0 >97 °C.

We identified two antimicrobial peptides (AMPs) with similarity to moronecidin in Antarctic fishes. The characteristics of both AMPs were determined using as a control. Moronecidin, which was first isolated from hybrid striped bass, is highly salt-resistant, and possesses broad-spectrum activity against various microbes. moronecidin-like peptide Notothenia coriiceps exhibited narrower spectrum higher salt sensitivity than moronecidin. AMP Parachaenichthys charcoti similar moronecidin,...

Cold-active acetyl xylan esterases allow for reduced bioreactor heating costs in bioenergy production. Here, we isolated and characterized a cold-active esterase (PbAcE) from the psychrophilic soil microbe Paenibacillus sp. R4. The enzyme hydrolyzes glucose penta-acetate acetate, reversibly producing xylan, it shows higher activity at 4°C than 25°C. We solved crystal structure of PbAcE 2.1-Å resolution to investigate its active site reason low-temperature activity. Structural analysis showed...

Parochlus steinenii is the only flying insect native to Antarctica. To elucidate molecular mechanisms underlying its adaptation cold environments, we conducted comparative genomic analyses of P. and closely related lineages. In an analysis gene family evolution, 68 rapidly evolving families, involved in innate immune system, unfolded protein response, DNA packaging, folding, unsaturated fatty acid biosynthesis were detected. Some families steinenii-specific showed phylogenetic instability....

Parochlus steinenii is a winged midge from King George Island. It cold-tolerant and endures the harsh Antarctic winter. Previously, we reported genome of this midge, but assembly with short reads had limited contig contiguity, which reduced completeness annotated gene sets. Recently, contiguity has been increased using nanopore technology. A number methods for enhancing low base quality have reported, including long-read (e.g. Nanopolish) or short-read Pilon) based methods. Based on these...
