Worldwide, myopia is the leading cause of visual impairment. It results from inappropriate extension ocular axis and concomitant declines in scleral strength thickness caused by extracellular matrix (ECM) remodeling. However, identities initiators signaling pathways that induce ECM remodeling are unknown. Here, we used single-cell RNA-sequencing to identify activated sclera during development. We found hypoxia-signaling, eIF2-signaling, mTOR-signaling were murine myopic sclera. Consistent...

Purpose.: To determine whether hypertonic stress promotes increases in inflammatory cytokine release through transient receptor potential vanilloid channel type 1 (TRPV1) signaling pathway activation human corneal epithelial cells (HCECs). Methods.: Hyperosmotic medium was prepared by supplementing isotonic Ringers solution with sucrose. Ca2+ measured fura2-AM–loaded HCECs using a single-cell fluorescence imaging system. Western blot analysis evaluated the phosphorylation status of EGFR,...

Purpose This study compares signs, symptoms and predictive tools used to diagnose dry eye disease (DED) ocular surface disorders in six systemic well-defined non-overlapping diseases. It is well known that these tests are problematic because of a lack agreement between them identifying conditions. Accordingly, we provide here comparative clinical profile analysis different Methods A spontaneous continuous sample patients with Sjögren's syndrome (SS) (n = 27), graft-versus-host-disease (GVHD)...

To determine whether dopamine receptor D1 (D1R) signaling pathway activation by bright light (BL) in specific retinal neuronal cell types contributes to inhibiting form-deprivation myopia (FDM) mice.Mice (3-weeks old) were raised under either normal (NL: 100-200 lux) or BL (2500-5000 conditions with without form deprivation. Refraction changes evaluated an eccentric infrared photorefractor, and ocular axial components optical coherence tomography. The D1R antagonist, SCH39166, was...

Sleep deficiency is a common public health problem associated with many diseases, such as obesity and cardiovascular disease. In this study, we established sleep deprivation (SD) mouse model using 'stick over water' method observed the effect of on ocular surface health. We found that SD decreased aqueous tear secretion; increased corneal epithelial cell defects, sensitivity, apoptosis; induced squamous metaplasia epithelium. These pathological changes mimic typical features dry eye....

In certain epithelial tissues, activation of transient receptor potential (TRP) vanilloid subtype 1 (TRPV1) by noxious stimuli induces pro-inflammatory cytokine release, which helps to mitigate the challenge. While corneal epithelium elicits such responses a variety challenges, it remains unknown whether TRPV1 mediates secretion. Accordingly, we probed for expression and function in human (HCEC) rabbit cell (RCEC) lines, their primary counterparts, mouse situ. Cell membrane perinuclear was...

Peroxisome proliferator-activated receptors (PPARs) are members of a superfamily nuclear transcription factors. They involved in mediating numerous physiological effects humans, including glucose and lipid metabolism. PPARα ligands effectively treat dyslipidemia have significant antiinflammatory anti-atherosclerotic activities. These their ligand-dependent activity make obvious targets for drug design. Here, we present the structure human complex with WY14643, member fibrate class drug,...

The peroxisome proliferator-activated receptor γ (PPARγ) is a target for treatment of type II diabetes and other conditions. PPARγ full agonists, such as thiazolidinediones (TZDs), are effective insulin sensitizers anti-inflammatory agents, but their use limited by adverse side effects. Luteolin flavonoid with actions that binds but, unlike TZDs, does not promote adipocyte differentiation. However, previous reports suggested variously luteolin agonist or an antagonist. We show exhibits weak...

Abstract N 6 ‐methyladenosine (m A) is a novel epitranscriptomic marker that contributes to regulating diverse biological processes through controlling messenger RNA metabolism. However, it unknown if m A methylation affects uveal melanoma (UM) development. To address this question, we probed its function and molecular mechanism in UM. Initially, demonstrated global levels were dramatically elevated both UM cell lines clinical specimens. Meanwhile, found METTL3, main regulatory enzyme, was...
