The microbial environment is an important factor that contributes to the pathogenesis of atopic dermatitis (AD). Recently, it was revealed not only bacteria itself but also extracellular vesicles (EVs) secreted from affect allergic inflammation process. However, almost all research carried out so far related local microorganisms, systemic distribution. We aimed compare bacterial EV composition between AD patients and healthy subjects experimentally find beneficial effect some composition.
We have identified an 8-gene signature with significant expression differences between gastric cancer and normal tissues. This set can predict the status of tissues more than 96% accuracy in a totally independent microarray dataset. The 8 genes are composed down-regulated KLF4, GPX3, SST LIPF, together up-regulated SERPINH1, THY1 INHBA cancer. To corroborate differential gene pattern, we chose GPX3 examined its pattern detail. A comparison shows broader multiple types cancers, including...
Although elevated syndecan-2 expression is known to be crucial for the tumorigenic activity in colon carcinoma cells, how regulates cancer unclear. In human adenocarcinoma tissue samples, we found that both mRNA and protein of were increased, compared with neighboring normal epithelium, suggesting plays functional roles cells. Consistent this notion, syndecan-2-overexpressing HT-29 cells showed enhanced migration/invasion, anchorage-independent growth, primary tumor formation nude mice,...
The Ni(II) complex of the tripeptide NH2-Gly-Gly-His-COOH is shown to mediate efficient protein-protein cross-linking in presence oxidants such as oxone and monoperoxyphthalic acid. Only proteins that associate specifically solution are cross-linked under these conditions. Preliminary probes mechanism reaction suggest active intermediate may be a high-valent metal attacks aromatic amino acids.
Resveratrol (3,5,4'-trihydroxy-trans-stilbene) is a natural compound found in large quantities, most notably grapes and red wine, which has been shown to have anti-inflammatory, chemopreventive anti-angiogenic effects. We examined whether resveratrol any effect on growth gene expression the human ovarian cancer PA-1 cells. show that inhibits cell induces apoptosis also investigated of changes global during resveratrol-induced inhibition cells using cDNA microarray with 7,448...
The mediator is an approximately 20 protein complex that essential for the transcription of most genes in yeast. It contacted by a number gene-specific activators, but details these interactions are not well understood cases. Here, evidence presented component Gal11 represents at least one target Gal4 activation domain (AD). Deletion shown to decrease affinity AD mediator, and direct binding N-terminal with demonstrated. Quantitative studies, however, indicate KD 1:1 AD−Gal11 modest....
There has been considerable interest in virulence genes the plasticity region of Helicobacter pylori , but little is known about many these genes. JHP940, one factors encoded by H. strain J99, a proinflammatory protein that induces tumor necrosis factor-alpha and interleukin-8 secretion as well enhanced translocation NF-κB cultured macrophages. Here we have characterized structure function JHP940 to provide framework for better understanding its role inflammation . Our work demonstrates...
Atopic dermatitis (AD) is an inflammatory skin disease, significantly affecting the quality of life. Using AD as a model system, we tested successive identification AD-associated microbes, followed by culture-independent serum detection identified microbe.
Purpose: Chitin is a potent adjuvant in the development of immune response to inhaled allergens airways.According other studies, chitin known as multi-faced adjuvants which can induce Th2 responses.Recently, we found that TNF-α key mediator cell allergens. Here, evaluated immunologic mechanisms airway hypersensitivity allergens, enhanced by house dust mite (HDM)-derived chitin. Methods: The role and TLRs was an mouse model induced sensitization with allergen (ovalbumin, OVA) HDM-derived using...
Repetitive exposure to ultraviolet B (UVB) radiation is known cause DNA damage and increase levels of reactive oxy-gen species (ROS), which are linked the upregulation matrix metallopeptidases (MMPs) inflammatory cyto-kines. These events lead collagen breakdown by MMPs, resulting in wrinkles, loss elasticity, rough, dry skin—typical signs photoaging—and may even contribute skin cancers long term. We have previously shown that Solanum tuberosum-Derived Exosomes (SDE) can repress 80%...
Background/Objectives: Potato (Solanum tuberosum)-derived exosomes (SDEs) are extracellular vesicles (66 nm in diameter) with therapeutic potential. SDEs suppress matrix metallopeptidases (MMPs) 1, 2, and 9, tumor necrosis factor (TNF), interleukin 6 (IL6), while exhibiting radical-scavenging activity against the free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH) vitro mitigating hydrogen peroxide (H2O2)-induced oxidative stress HaCaT cells. upregulate antioxidant gene glutathione...
Abstract Aberrant constitutive expression of the NF‐κB c‐Rel and RelA subunits in breast cancer cells was shown to promote their survival. Recently, we demonstrated that aggressive cancers constitutively express high levels RelB subunit, which promotes more invasive phenotype via induction BCL2 gene. As these are frequently resistant therapy, here tested hypothesis High expressing Hs578T MDA‐MB‐231 were γ‐radiation than MCF7 ZR‐75 cells, lower levels. Knockdown led decreased survival...
There are hump, humpless cattle and gayal distributed in Yunnan province, south-west China, but their genetic background remains unclear. To determine the origin diversity of (Diqing, Nujiang Wenshan cattle), we analysed mtDNA control region sequences 71 samples SRY gene 39 samples, together with available GenBank. The neighbour-joining phylogeny reduced median network analysis showed that originated from hybridization between male Bos frontalis female taurus or indicus, mostly B. also...
Accumulating evidence has shown that the Toll-like receptor 7 agonist imiquimod (IMQ) induces psoriasiform skin inflammation in mice and this is dependent on IL-23/IL-17 axis. Moreover, it been demonstrated main source of IL-17 not Th17 but dermal gamma delta (γδ) T cells mouse skin. Recent advances understanding immunopathogenesis psoriasis led to an alteration treatment paradigm use highly efficacious biologics. However, their high cost impedes extensive these agents. Thus, inexpensive...
