Abstract Gain-of-function mutations in the human SCN11A -encoded voltage-gated Na + channel V 1.9 cause severe pain disorders ranging from neuropathic to congenital insensitivity. However, entire spectrum of diseases has yet be defined. Applying whole-exome sequencing we here identify a missense change (p.V1184A) 1.9, which leads cold-aggravated peripheral humans. Electrophysiological analysis reveals that p.V1184A shifts voltage dependence opening hyperpolarized potentials thereby...
We report a new class of deubiquitinating enzyme (DUB) probes that resemble the native diubiquitin with same linkage size and contain Michael addition acceptor for trapping DUB active-site cysteine. Both K63- K48-linked were generated using facile chemical ligation method. The diUb demonstrated to label DUBs from different families revealed intrinsic specificities DUBs.
Abstract To analyze the intestinal microbiota diversity of several important economic fishes in Loudi area and its correlation with water environment, high-throughput sequencing based on bacteria 16S rRNA was used to fish intestines water. The results revealed that half OTUs sample could be detected intestine, proportion shared Hypophthalmichthys molitrix samples only 22%, unique OTU LC group relatively highest group. It can seen from analysis NMDS analysis, distance between is farthest....
As a popular tool for gene function characterization and therapy, RNA interference (RNAi)-based silencing has been increasingly explored potential applications to control invasive species. At least two major hurdles exist when applying this approach plants: (1) the design screening of species- gene-specific biomacromolecules (i.e., gene-silencing agents or GSAs) made DNA, RNA, peptides that can suppress expression target genes efficiently, (2) delivery vehicle needed penetrate plant cell...
Cervical cancer is one of the cancers commonly found in female reproductive system and associated with obesity. However, exact connection mechanisms remain unclear. Screening key therapeutic targets natural products good anti-tumor activity has become a crucial strategy for therapy. Cryptotanshinone anti-inflammatory anti-cancer properties. Key related low-toxicity active ingredients were identified as components treatment strategies. Therefore, network pharmacology cellular biology...
DNA damage response (DDR) is crucial for maintaining genomic stability and preventing the accumulation of mutations that can lead to various diseases, including cancer. DDR a complex cellular regulatory network involves sensing, signal transduction, repair, cell cycle arrest. Modifications in histone phosphorylation play important roles these processes, facilitating repair factor recruitment, chromatin remodeling, regulation. The precise regulation critical effective damage, integrity...
Synthetic non-metallic nanoparticles (NMNPs) such as carbon nanotubes (CNTs), nanodots (CNDs), and cell-penetrating peptides (CPPs) have been explored to treat harmful algal blooms. However, their strain-specific algicidal activities rarely investigated. Here we determined acute toxicity nine freshwater cyanobacterial strains belonging seven genera, including Microcystis aeruginosa UTEX 2386, M. 2385, LE3, Anabaena cylindrica PCC 7122, Aphanizomenon sp. NZ, Planktothrix agardhii SB 1810,...
Abstract Huntington disease (HD) is an autosomal dominant, progressive neurodegenerative disorder for which there no cure. Predictive testing HD available to asymptomatic at‐risk individuals. Approximately half of the population undergoing predictive consists young adults (≤35 years old). Finishing one's education, starting a career, engaging in romantic relationships and becoming parent are key milestones adulthood. We conducted qualitative study explore how gene‐positive influences adults’...
Activity-based Ub-PCNA probes identify deubiquitinating enzymes (DUBs) specific for PCNA and demonstrate site-specific deubiquitination by DUBs.
Polyubiquitination of proteins regulates a variety cellular processes, including protein degradation, NF-κB pathway activation, apoptosis, and DNA damage tolerance. Methods for generating polyubiquitinated with defined ubiquitin chain linkage length are needed an in-depth molecular understanding polyubiquitination. However, enzymatic polyubiquitination usually generates mixed lengths in low yield. We report herein new chemical approach under mild condition that preserves the native fold...
Abstract Background Barramundi ( Lates calcarifer ) is an important farmed marine food fish species. Its first generation linkage map has been applied to QTL for growth traits. To identify genes located in responsible specific traits, genomic large insert libraries are of crucial importance. We reported herein a bacterial artificial chromosome (BAC) library and the mapping BAC clones map. Results This consisted 49,152 with average size 98 kb, representing 6.9-fold haploid genome coverage....
We report a cysteine-based ligation strategy for generating monoubiquitylated protein while preserving the native cysteine residues on acceptor protein. In monoubiquitylation of proliferating cell nuclear antigen (PCNA) this method circumvents need to mutate PCNA. The chemically ubiquitylated PCNA contains noncleavable linkage same length as isopeptide linkage. It also retains normal function Ub-PCNA in stimulating ATPase activity replication factor C (RFC) and lesion bypass synthesis by...
Deubiquitination has emerged as an essential regulatory mechanism of a number cellular processes. An in-depth understanding deubiquitinating enzyme (DUB) catalysis, particularly the mode ubiquitin binding and individual steps in DUB catalytic turnover, is imperative for exploiting DUBs therapeutic intervention. In this work, we present transient kinetic study USP2 hydrolyzing model substrate Ub-AMC physiological K48-linked diubiquitin. We conducted stopped-flow fluorescence analyses mono-...
C. testosteroni is a research topic that can degrade steroid hormones into water and carbon dioxide through series of enzymes in the body. Short-chain dehydrogenase (SDR) are class NAD (P) H-dependent oxidoreductases testosteroni. Its main function catalyzing redox hydroxyl/ketone group hormone. In this paper, SDR gene(SDRx) cloned from ATCC11996 expressed. The polyclonal antibody was prepared SDRx gene knocked out by homologous recombination. Wild type mutant induced testosterone,...
The photophysical properties of four Ir(III) complexes have been investigated by means the density functional theory/time-dependent theory (DFT/TDDFT). effect electron-withdrawing and electron-donating substituents on charge injection, transport, absorption phosphorescent has studied. theoretical calculation shows that lowest-lying singlet absorptions for 1–4 are located at 387, 385, 418 386 nm, respectively. For 1–4, phosphorescence 465, 485, 494 478 nm is mainly attributed to LUMO → HOMO...
