Efficacy and tolerability of the new antiepileptic drug oxcarbazepine, was evaluated in a retrospective multicentre study. The records all 947 epilepsy patients treated with oxcarbazepine eight participating centres from 1981 through 1990 were examined. median daily dose 30 mg/kg children, 18 adults, 15 elderly patients, given b.i.d. or t.i.d. mean plasma levels main active metabolite 88, 79, 68 mumol/l elderly, respectively. In shifted to treatment, seizure frequency unchanged 51-66%,...

DNA-encoded small molecule libraries (DELs) have facilitated the discovery of novel modulators many different therapeutic protein targets. We report first successful screening a multimillion membered DEL inside living cell. demonstrate method using oocytes from South African clawed frog Xenopus laevis. The large size 1 μL, or 100 000 times bigger than normal somatic cell, permits simple injection DELs, thus resolving fundamental problem delivering DELs across cell membranes for in vivo...

Limited solubility and precipitation of amyloidogenic sequences such as the Alzheimer peptide (β-AP) are major obstacles to a molecular understanding protein fibrillation deposition processes. Here we have circumvented problem by stepwise engineering β-AP homology into soluble scaffold, monomeric S6. The S6 construct with highest crystallizes tetramer that is linked residues forming intermolecular antiparallel β-sheets. This also shows increased coil aggregation during refolding, 14-mer...

The interpretation of folding rates is often rationalized within the context transition state theory. This means that reaction rate linked to an activation barrier, height which determined by free energy difference between a ground (the starting point) and apparent state. Changes in kinetics are thus caused effects on either state, or both. However, structural changes rarely discussed connection with experimental data, kinetic anomalies commonly ascribed alone, e.g., depletion accumulation...

Ras GTPase Activating Protein SH3 Domain Binding (G3BP) is a potential anti-cancer drug target implicated in several cellular functions. We have used protein crystallography to solve crystal structures of the human G3BP1 NTF2-like domain both alone and complex with an FxFG Nup repeat peptide. Despite high structural similarity, binding site located between two alpha helices not at dimer interface as observed for nuclear transport factor 2. ITC studies showed specificity towards motif but FG...
