We studied the effects of endothelium‐derived relaxing factor (EDRF), bovine retractor penis muscle inhibitory and sodium nitroprusside, three stimulants guanylate cyclase, on in vitro aggregation washed human platelets. Platelet induced either by collagen or thromboxane A 2 analogue U46619 was inhibited all agents. The anti‐aggregatory effect each agent haemoglobin. EDRF potentiated superoxide dismutase. These findings are discussed relation to a potential role for haemostasis.
1. In isolated heart muscle preparations, selective removal of the endocardium results in a characteristic and unusual negative inotropic effect. Possible mechanisms for this effect were investigated study. 2. endocardium‐intact preparations ferret papillary muscle, 8‐bromo‐cyclic GMP, sodium nitroprusside, atrial natriuretic peptide (ANP) substance P each induced changes contractile behaviour similar to endocardial removal, significantly elevated myocardial cyclic GMP levels. Substance...
The mechanisms by which haemoglobin and methaemoglobin inhibit the vasodilator actions of glyceryl trinitrate, sodium azide, nitric oxide, bovine retractor penis inhibitory factor (IF) were studied on rabbit endothelium‐denuded aortic rings. Methaemoglobin was less effective than against each vasodilator, it more at inhibiting relaxation to azide that trinitrate. Glyceryl trinitrate neither bound nor inactivated when passed through columns haemoglobin‐agarose or methaemoglobin‐agarose. Azide...
Endocardial cells release factors which regulate myocardial contractility and guanosine 3′:5′‐cyclic monophosphate (cyclic GMP) levels. One of these is indistinguishable from endothelium‐derived relaxing factor (EDRF). The effluent pig heart endocardial cultured on microcarrier beads caused the relaxation a coronary artery ring denuded endothelium. This was enhanced by combination superoxide dismutase catalase attenuated haemoglobin, binds nitric oxide (NO), inhibitors NO synthase, N G...
Human papillomavirus (HPV) type 16 is implicated in the aetiology of anogenital dysplasia which may progress to malignancy. HPV-16 DNA actively transcribed cervical carcinomas, most abundant transcripts being from E6 and E7 early open reading frames. The protein has been shown have transforming activity vitro. In this report we define four immunodominant B epitopes within corresponding gene, using a panel murine monoclonal antibodies. Three are linear lie N-terminal region molecule, unique...
We have investigated whether porcine endocardial cells in culture express the inducible, Ca 2+ ‐independent form of nitric oxide (NO) synthase. NO synthase activity cytosolic extracts was measured by estimation rate formation l ‐[ 14 C]‐citrulline from C]‐arginine. Treatment with lipopolysaccharide or cytokines induced a cell cytosol. The combination tumour necrosis factor (TNFα, 10 ng ml −1 ) and interleukin‐1β (IL‐1β, greatest enzyme activity. increased following exposure to parallelled an...
1. Unactivated extracts of bovine retractor penis (BRP) contains 3-7 microM nitrite. Acid-activation these at pH 2 for 10 min followed by neutralization generates the active form inhibitory factor (IF; assayed its vasodilator action on rabbit aorta), and is associated with partial loss 2. Increasing time acid-activation from to 60 intermittent vortex mixing greater activity increases nitrite loss. 3. When acid-activated neutralized are incubated 37 degrees C or 30 boiled 5 min, lost content...
Functional analysis of mutant class II major histocompatibility complex molecules has begun to identify regions important for antibody binding and T-cell activation. By using in vitro mutagenesis directed at the beta 1 domain Ab gene we have constructed three structurally distinct genes. Each these genes, as well wild-type gene, was cotransfected together with alpha into Ia-negative B-lymphoma cell line M12.C3. Transfection resulted successful synthesis surface expression antigens that...
Cultured aortic endothelial cells of the pig respond to endothelium‐derived relaxing factor (EDRF) they release with an increase in cyclic GMP content. This response is inhibited by haemoglobin or l ‐N G ‐monomethyl‐arginine ( ‐NMMA), and has been used investigate effects phorbol esters on EDRF release. Pretreatment phorbol‐12,13‐dibutyrate (PDB) but not inactive 4α‐phorbol‐12,13,‐didecanoate (PDD), increases induced substance P (10 −8 m ) a time concentration‐dependent manner. PDB did...
Insulin is a major autoantigen in type 1 diabetes, targeted by both CD8 and CD4 T cells. We studied an insulin-reactive T-cell receptor (TCR) α-chain transgenic NOD mouse on TCRCα proinsulin 2 (PI2)-deficient background, designated as A22Cα−/−PI2−/− mice. These mice develop low incidence of autoimmune diabetes. To test the role gut microbiota diabetes development this model system, we treated with enrofloxacin, broad-spectrum antibiotic. The treatment led to male developing accelerated found...
Endothelium‐dependent and ‐independent relaxation of aortic ring preparations was assessed nitric oxide (NO) synthase activity measured in the lung, cerebellum cholesterol‐fed normal rabbits. acetylcholine ATP depressed while that to calcium ionophore, A23187, unaltered group. Relaxation sodium nitroprusside however greater aortae from animals. Neither Ca 2+ ‐dependent nor NO could be detected or hearts taken either group Activity both enzymes cerebellae groups enzyme significantly (ca. 2...
Summary: Endothelial cells are known to contain both soluble and particulate guanylate cyclase, but the functional role of cyclic guanosine monophosphate (cGMP) in endothelial remains unknown. We have investigated effects 8-bromo-cGMP on cndothelium-dependent relaxations acetylcholine, substance P, ATP, calcium ionophore A23I87, endothelium-indepen-dent sodium nitroprusside glyceryl trinitrate (GTN). The ability each these agents relax phenylephrine-preconstricted rings rabbit aorta was...
A panel of murine mAb raised against a MS2 replicase/HPV 18 E7 fusion protein included 23 reactive by ELISA with HPV determinants. total 19 the recognized linear epitopes in N-terminal region molecule, while other four were deduced binding inhibition assays to recognize conformational determinants this region. All tested antibodies precipitated 14-kDa peptide doublet that corresponded predicted size protein, from HeLa cells, but not 16 containing CaSki cells. was detected immunolabeling...
We examined the effects of haemoglobin (which inhibits vascular responses to stimulation soluble guanylate cyclase) and M&B 22,948 selectively cyclic GMP phosphodiesterase) on relaxation induced in rabbit aorta by atrial natriuretic peptide, atriopeptin II stimulates particulate cyclase). Pretreatment with (100 microM) produced a 2.3 fold potentiation II-induced endothelium-denuded rings aorta. (10 had no effect or 10.9 increase content The suggests causal role for mediating vasodilatation...
Abstract Light and smell have both been shown to induce beneficial changes human psychophysiology. Bright light therapy has a positive impact on anxiety depression also effects mood, stress, depression. We developed method for the delivery of integrated stimulation try optimise psychophysiological benefit. tested its effectiveness physiological measure, EEG frontal alpha asymmetry (FA) psychological paradigm, POMS test, which used as measure emotional state mood. Light, pleasant smell,...
Summary: The endocardium of isolated cat cardiac papillary muscle preparations has recently been reported to modulate myocardial contraction. We investigated the effects selective removal (by a 2-s immersion in 0.5% Triton X-100) on contraction ferret muscles (n = 15). Endocardium resulted characteristic immediate abbreviation twitch with small reduction peak contractile performance but no significant change unloaded shortening velocity. not duration were calcium dependent. These...
