Curcumin has been demonstrated to have many neuroprotective properties, including improvement of cognition in humans and neurogenesis animals, yet the mechanism such effects remains unclear. We assessed behavioural performance hippocampal cell proliferation aged rats after 6- 12-week curcumin-fortified diets. enhanced non-spatial spatial memory, as well dentate gyrate compared control diet rats. We also investigated underlying mechanistic pathways that might link curcumin treatment increased...

Abstract Alzheimer’s disease (AD) is a common neurodegenerative characterized clinically by progressive deterioration of memory, and pathologically histopathological changes including extracellular deposits amyloid-beta (A-beta) peptides forming senile plaques (SP) the intracellular neurofibrillary tangles (NFT) hyperphosphorylated tau in brain. This review focused on new developments amyloid cascade hypothesis with details production, metabolism clearance A-beta, key roles some important...

PI3K-Akt-mTOR signaling pathway has been validated as an effective targeting for cancer therapy. However, no PI3K/mTOR dual inhibitor approved by the FDA yet. Therefore, it is still essential to discover a candidate with good efficacy and low toxicity. In our design, series of imidazo[1,2-a]pyridine derivatives had synthesized subjected activity assessment in vitro vivo. 15a was proved be potent excellent kinase selectivity, modest plasma clearance, acceptable oral bioavailability. Besides,...

Abstract Alkene hydrodifluoroalkylation is a fruitful strategy for synthesizing difluoromethylated compounds that are interesting developing new medicinal agents, agrochemicals, and advanced materials. Whereas the anti-Markovnikov to linear-type products developed, employing radical-based processes, Markovnikov synthesis of branched adducts remains unexplored. Herein, we describe acid-catalyzed processes involving carbocation intermediates as promising secure regioselectivity. Accordingly,...

In addition to senile plaques and cerebral amyloid angiopathy, the hyperphosphorylation of tau protein formation intraneuronal neurofibrillary tangles (NFTs) represents another neuropathological hallmark in AD brain. Tau is a microtubule-associated localizes predominantly axons neurons with primary function maintaining microtubules stability. When balance between phosphorylation dephosphorylation changed favor former, hyperphosphorylated level free fractions elevated. The NFTs represent...

Abstract Presenilin (PS) 1 and 2 conditional double knockout (cDKO) mice show progressive memory dysfunction forebrain degeneration. Gene expression profiling results revealed a strong activation of immunity inflammation responses in the brains 10‐month‐old cDKO mice. As environmental enrichment (EE) has been shown to be able improve induce neurogenesis brain, we assessed effects EE on performance neurodegeneration We found that effectively enhanced partially rescued atrophy Our suggest...

The C(sp2)–H function of indole ketone with diazo compound via a rhodium(II)-catalyzed intramolecular electrophilic trapping reaction under mild conditions in air was demonstrated. established methodology provided highly efficient approach for direct synthesis mutisubstituted tetrahydrocarbazoles continuous quaternary carbons. resulting products facilitate further modification to conveniently construct additional fused heterocyclic rings. By phenotypic screening, several exhibit good...

Mitochondrial serine hydroxymethyl transferase isoform 2 (SHMT2) has attracted increasing attention as a pivotal catalyzing regulator of the serine/glycine pathway in one-carbon metabolism cancer cells. However, few inhibitors that target this potential anticancer have been discovered. Quantitative characterization interactions between SHMT2 and its known should benefit future discovery novel inhibitors. In study, we employed recently developed alanine-scanning-interaction-entropy method to...

Human mitochondrial peptide deformylase (HsPDF) is responsible for removing the formyl group from N-terminal formylmethionines of newly synthesized proteins and plays important roles in maintaining mitochondria function. It overexpressed various cancers has been proposed as a novel therapeutic target. Actinonin, naturally occurring peptidomimetic HsPDF inhibitor, was reported to inhibit proliferation broad spectrum human cancer cells vitro. However, its efficacy pharmacokinetic profile...

We report an efficient Au(I)-catalyzed formal allylation of diazo(thio)oxindoles using allyltrimethylsilane to give 3-allyl (thio)oxindoles, which are difficult access by traditional alkylation methods under basic conditions. The approach enables a highly stereoselective synthesis quaternary (thio)oxindoles via allylation-asymmetric Michael addition sequence. These adducts versatile synthons for spirocyclic (thio)oxindoles. Initial biological studies reveal that chiral thiooxindoles show...

Pyrrolidines 3h 3k showed good proliferation inhibitory effects toward 10 cancer cell lines with IC 50 values ranging from 2.9 to 16 μM. Furthermore, pyrrolidine induced cycle arrest at the G0/G1 phase time‐ dose‐dependent cellular apoptosis in both HCT116...
