In autophagy, the double-membrane autophagosome delivers cellular components for their degradation in lysosome. The conserved Ypt/Rab GTPases regulate all trafficking pathways, including autophagy. These function modules that include guanine-nucleotide exchange factor (GEF) activators and downstream effectors. Rab7 its yeast homologue, Ypt7, context of such a module, fusion both late endosomes autophagosomes with yeast, Rab5-related Vps21 is known role early- to late-endosome transport. Here...

Nitrogen starvation is a universal stimulus of autophagy. At present, little known about the relationship between carbon metabolism and autophagy under nitrogen starvation. Here, we show that yeast cells continue to consume glucose downregulate fermentation Storage lipid production increased, with concurrent proliferation droplets. Furthermore, provide evidence triacylglycerol synthesis crucial for autophagosome biogenesis. It involved in step downstream PAS (phagophore assembly site)...

Triple-negative breast cancer (TNBC) is the leading cause of cancer-related death in women. Previous studies indicated that miR-361-5p was downregulated cancer, however, exact effect on TNBC requires further investigation. In present study, we investigated whether can act as a tumor suppressor by targeting required for cell differentiation 1 homolog (RQCD1) and inhibiting epidermal growth factor receptor (EGFR)/phosphoinositide 3-kinase (PI3K)/protein kinase B (Akt) pathway TNBC. The...

Eukaryotic cells share a basic scheme of internal organization featuring membrane-based organelles. The use fluorescent proteins (FPs) greatly facilitated live-cell imaging organelle dynamics and protein trafficking. One major limitation this approach is that the fusion an FP to target can often does compromise function alter its subcellular localization. optimization process obtain desirable construct be time-consuming or even unsuccessful. In work, we set out provide validated FP-based...

Fluorescence microscopy of live cells is instrumental in deciphering the molecular details autophagy. To facilitate routine examination yeast Atg proteins under diverse conditions, here we provide a comprehensive tool set, including (1) plasmids for expression GFP chimeras at endogenous levels most proteins, (2) RFP-Atg8 constructs with improved properties as PAS marker, and (3) complementation common auxotrophic markers. We hope that availability this set will further accelerate autophagy research.

The formation of autophagosomes mediating the sequestration cytoplasmic materials is central step autophagy. Several phosphoinositides, which are signaling molecules on membrane, involved in However, it not always clear whether these phosphoinositides act directly at site autophagosome formation, or indirectly via regulation other steps pathways. To address this question, we used a set phosphoinositide probes to systematically examine their potential presence autophagosomal membranes yeast...

In Saccharomyces cerevisiae, Atg9 is an important autophagy-related (Atg) protein, and interacts with hundreds of other proteins. How many Atg9-interacting proteins are involved in macroautophagy/autophagy unclear. Here, we conducted a multi-omic profiling Atg9-dependent molecular landscapes during nitrogen starvation-induced autophagy, identified 290 256 genes to be markedly regulated by ATG9 transcriptional translational levels, respectively. Unexpectedly, found most known Atg autophagy...

ABSTRACT For Saccharomyces cerevisiae , gene knockout is routinely performed by transformation with a linear DNA cassette consisting of selection marker flanked upstream and downstream sequences homologous to target gene. Over the years, several plasmid sets containing variety genes have been developed. Targeting fidelity under this strategy was high when performing first in strain. However, we found that targeting decreased substantially subsequent knockouts. The majority transformants were...
