ABSTRACT Colistin and polymyxin B remain part of the last line antibiotics for multidrug-resistant Gram-negative bacteria, such as carbapenem-resistant Enterobacteriaceae . Current joint EUCAST-CLSI recommendations are broth microdilution (BMD) to be performed MIC testing colistin. Commercial susceptibility methods were evaluated compared against reference BMD, using a breakpoint ≤2 mg/liter both colistin B. Seventy-six included, which 21 mcr-1 positive (18 Escherichia coli isolates, 2...

Since 2010, the incidence of carbapenem-resistant Enterobacteriaceae (CRE) has been increasing in Singapore. We analyzed clinical and molecular epidemiology CRE among adult inpatients Singapore.Quarterly unique subjects (per 100000 patient-days) with positive surveillance cultures for were estimated based on mandatory data submitted to National Public Health Laboratory by public hospitals between 2010 2015. CRE-positive prospectively recruited from 6 sector December 2013 April Subjects...

Owing to gene transposition and plasmid conjugation, New Delhi metallo-β-lactamase (NDM) is typically identified among varied Enterobacteriaceae species STs. We used WGS characterize the chromosomal molecular epidemiology of NDM transmission involving four institutions in Singapore. Thirty-three isolates (collection years 2010–14) were sequenced using short-read sequencing-by-synthesis analysed. Long-read single molecule, real-time sequencing (SMRTS) was genetically a novel pSg1-NDM carried...

In this study, we describe the characterization of an infrequently encountered class A carbapenemase, IMI-1, from a clinical Enterobacter cloacae isolate. The isolate had high levels resistance to carbapenems but retained susceptibility expanded-spectrum cephalosporins. bla IMI-1 gene was chromosomally encoded. Detection producer highlights diversity carbapenemases in local setting.

Tropheryma whipplei is a poorly studied bacterium responsible for Whipple's disease. In this study, its susceptibility to doxycycline was investigated at transcriptional level using whole-genome DNA microarray.Exposure of T. the MIC (0.5 mg/L) induced antibiotic-specific primary expression profiles, while indirect effects were detected 10 x MIC. contrast what observed several microorganisms exposed antibiotics, heat-shock proteins not affected. Abstract Background The aim of this study was to analyze the genomic diversity several Tropheryma whipplei strains by microarray-based comparative hybridization. Fifteen clinical isolates originating from biopsy samples recovered different countries were compared with T. Twist strain. For each isolate, genes defined as either present or absent/divergent using GACK analysis software. Genomic changes then further characterized PCR and sequencing. Results results revealed a limited genetic...

Whole-genome sequencing was performed on 16 isolates of the carbapenemase-producing Enterobacter cloacae complex to determine flanking regions blaIMI-type genes. Phylogenetic analysis multilocus sequence typing (MLST) targets separated into 4 clusters. The genes were all found Xer-dependent integrative mobile elements (IMEX). IMEX 5 similar those described in Canada, while remainder novel. Five had lacking a resolvase and recombinase.
