Significance The COVID-19 pandemic has already caused over 1 million deaths. Therefore, effective vaccine concepts are urgently needed. In search of such a concept, we have analyzed measles virus-based candidate targeting SARS-CoV-2. Using this well-known, safe backbone, demonstrate here induction functional immune responses in both arms adaptive immunity, yielding antiviral efficacy vivo with the desired bias. Consequently, no immunopathologies became evident during challenge experiments....
Vaccine-associated enhanced respiratory disease (VAERD) is a severe complication for some infections. To investigate the potential VAERD induction in coronavirus 2019 (COVID-19), we evaluate two vaccine leads utilizing hamster infection model: T helper type 1 (TH1)-biased measles vaccine-derived candidate and TH2-biased alum-adjuvanted, non-stabilized spike protein. The virus (MeV)-derived protects animals, but protein lead induces VAERD, which can be alleviated by dexamethasone treatment....
Vibrio parahaemolyticus exists as swimmer and swarmer cells, specialized for growth in liquid on solid environments respectively. Swarmer cells are characteristically highly elongated due to an inhibition of cell division, but still need divide order proliferate expand the colony. It is unknown how long without diminishing population required swarming behavior. Here we show that placement division site length-dependent; short swarmers at mid-cell, while switch a specific non-mid-cell site....
Abstract The COVID-19 pandemic is caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and has spread world-wide with millions of cases hundreds thousands deaths to date. gravity the situation mandates accelerated efforts identify safe effective vaccines. Here, we generated measles virus (MeV)-based vaccine candidates expressing SARS-CoV-2 spike glycoprotein (S). Insertion full-length S protein gene in two different MeV genomic positions resulted modulated expression....
The ability to introduce targeted genetic modifications in microbial genomes has revolutionized our study the role and mode of action individual bacterial virulence factors. Although fastidious lifestyle obligate intracellular pathogens poses a technical challenge such manipulations, last decade produced significant advances conduct molecular analysis Chlamydia trachomatis, major agent infertility blindness. Similar approaches have not been established for closely related veterinary spp.,...
The bacterial pathogen Chlamydia trachomatis employs the effector CpoS to suppress a host defense response that aborts intracellular growth by inducing cell death. While conducting CRISPR knock-out screen for genes contributing this response, we uncovered mutant deficient display markedly increased reliance on cellular ceramide synthesis, compensating its diminished ability acquire sphingolipids via modulating membrane trafficking. Employing power of just recently established molecular...
ABSTRACT Since December 2019, the novel human coronavirus SARS-CoV-2 has spread globally, causing millions of deaths. Unprecedented efforts have enabled development and authorization a range vaccines, which reduce transmission rates confer protection against associated disease COVID-19. These vaccines are conceptually diverse, including e.g. classical adjuvanted whole-inactivated virus, viral vectors, mRNA vaccines. We analysed two prototypic model strongly T H 1-biased measles...
