A "shot in the arm" for cancer therapy: Coupling of synthetic glycopeptide tandem-repeat sequences epithelial mucin MUC1 with Thomsen–Friedenreich (T) antigen (A) or a difluoro analogue (B) to tetanus toxoid (TTox) affords vaccines, which induce very strong immune responses mice overriding natural tolerance system. The induced antibodies are selectively directed against tumor-associated structures and strongly bind breast cells MCF-7 cell line.
An ideal combination: The vaccine A is obtained by conjugation of the synthetic tumor-associated sialyl-TN-MUC1 glycopeptide antigen to tetanus toxoid (molecular weight >150 000). Vaccine induces a very strong and selective immune response against structure in wild-type mice (Balb/c).
Objective To evaluate the influence of methylxanthine derivative, pentoxifylline, on plasma levels tumor necrosis factor (TNF)-alpha, interleukin (IL)-1, and IL-6 in prematurely delivered infants with generalized bacterial infections to assess effect this immunomodulating drug clinical outcome newborns sepsis. Design A prospective, randomized, double-blind trial. Setting The neonatal intensive therapy units university teaching hospitals. Patients One hundred patients sepsis admitted during a...
A shot in the arm for cancer treatment: Two MUC1 tetanus toxoid vaccines were synthesized and induced a strong immune response mice. The antibodies elicited by show high selectivity tumor cells mammary carcinoma tissues also distinguish between at different stages. ABSTRACT The assessment of intracellular cytokines at the single-cell level by flow cytometry has recently become a potent tool in many areas cell biology and defining role various human diseases. Three-color for detection combined with simultaneous determination lymphocytes (CD3 + CD4 ) or monocytes (CD33 CD14 was used comparison phytohemagglutinin (PHA)-and phorbol myristate acetate (PMA)-ionomycin-induced production peripheral blood mononuclear cells (PBMCs) healthy donors. We found that...
Sweet medicine: The mucin MUC1 is highly overexpressed on epithelial tumor cells. Glycopeptides 1 (from the repeating unit of MUC1) carrying various important sialylated tumor-associated carbohydrate antigens R were synthesized and coupled to bovine serum albumin (to generate anticancer vaccines) microtiter plates test selectivity immune response).
Mendelian susceptibility to mycobacterial diseases (MSMD) is a rare syndrome, the known genetic etiologies of which impair production of, or response interferon-gamma (IFN-γ). We report here patient (P1) with MSMD whose cells display mildly impaired responses IFN-γ, at levels, however, similar those from patients autosomal recessive (AR) partial IFN-γR2 STAT1 deficiency. Whole-exome sequencing (WES) and Sanger revealed only one candidate variation for both MSMD-causing IFN-γ-related genes....
In an attempt to elaborate in vitro on a therapeutic strategy that counteracts inflammatory signal, we previously reported novel immunopharmacological potential of glutathione, antioxidant thiol, regulating cytokines. the present study, investigated hypothesis selective regulation phosphodiesterases (PDEs), family enzymes controls intracellular cAMP/cGMP degradation, differentially regulates proinflammatory Selective PDE1 inhibition (8-methoxymethyl-3-isobutyl-1-methylxanthine) blockaded...
Tetrasubstituted pyrroles can be synthesized in a one-pot procedure from isoxazoles. The process includes the photoinduced situ formation of acylazirines combined with subsequent cobalt(II)-catalyzed ring expansion 1,3-diketones.
Block glycosylation of a T antigen–threonine conjugate with sialyl Lewisx trichloroacetimidate was used to synthesize Lewisx–T building block. Selective protecting-group manipulations furnished compound exclusively acetyl-protected functions in the carbohydrate portion; this intermediate is sufficiently acid-stable that it applicable solid-phase synthesis binding site A P-selectin ligand PSGL-1.
In this report we investigated the immunopharmacological role of selective and nonselective phosphodiesterase (PDE) inhibition in regulating inhibitory-kappaB (IkappaB-alpha)/nuclear factor-kappaB (NF-kappaB) signaling transduction pathway. fetal alveolar type II epithelial cells, PDE blockade at level diverging cAMP/cGMP pathways differentially regulated phosphorylation degradation IkappaB-alpha, major cytosolic inhibitor NF-kappaB. Whereas PDEs 1, 3, 4, by action...
Cooperation is key: Peptide and saccharide portions cooperate in sialyl Lewisx glycopeptides their mimetics the fucose sialic acid parts (see formula), resulting up to greater than 100-fold stronger binding E-selectin. The synthesis provided amino acids a form sufficiently acid-stable for application automated solid-phase syntheses of based on acid-sensitive linkers.
Total synthesis through block glycosylation and selective chemical O-sulfation of tyrosine residues yielded the glycopeptide recognition domain A (X=SO(3) (-)) P-selectin glycoprotein ligand 1, in which terminal sialic acid complex hexasaccharide side chain was replaced by (S)-cyclohexyl lactic acid. In binding assays O-sulfated structure showed high affinity towards P-selectin, non-sulfated E-selectin.
The selective C-terminal deprotection of O-glycopeptide (methoxyethoxy)ethyl esters is achieved under mild conditions (pH 6.6, 37 °C) by enzymatic hydrolysis using papain or lipase M from Mucor javanicus to give building blocks useful for chain-extending glycopeptide synthesis. On the other hand, removal acetyl protecting groups saccharide portion glycopeptides accomplished alternative with WG wheat germ furnish model substrates glycosyl transfer reactions in order extend carbohydrate side...
Abstract Intracellular expression of several cytokines was assessed in lymphocytes and monocytes children with transient hypogammaglobulinaemia infancy (THI) selective IgA deficiency (SIgAD). THI characterized by an increased frequency CD3+/CD4+ expressing tumour necrosis factor α (TNF-α), TNF-β interleukin 10 (IL-10), while SIgAD elevated numbers these cells containing TNF-α interferon γ (IFN-γ) were observed. No changes the number CD4+ T IL-4 both diseases noted. The proportion CD33+...
Human peripheral blood monocytes pretreated with human recombinant tumour necrosis factor alpha (rTNF) showed an enhanced ability to present a soluble antigen, purified protein derivate of tuberculin, autologous T lymphocytes as assessed by their increased proliferation in vitro. This enhancing activity was due TNF and not impurities preparations anti-TNF antibodies abolished this phenomenon. The rTNF-treated expression HLA-DR molecules co-stimulatory the murine thymocyte assay. Pretreatment...
