ObjectivesThe emerging fungal pathogens comprising the Candida haemulonii complex (Candida haemulonii, var. vulnera and duobushaemulonii) are notable for their antifungal resistance. Twelve isolates with phenotypic similarity to C. were recovered from patients in Brazilian hospitals. Here we aimed identify these by a molecular approach, using current classification of this complex, evaluate susceptibility profiles.
We present herein an improved assay for detecting the presence of extracellular proteases from microorganisms on agar plates. Using different substrates (gelatin, BSA, hemoglobin) incorporated into and varying culture medium composition, we were able to detect proteolytic activities Pseudomonas aeruginosa, Micrococcus luteus Serratia marcescens as well influence that these components displayed in expression enzymes. For all tested found agar-BHI or yeast extract containing gelatin...
Candida haemulonii, haemulonii var. vulnera and duobushaemulonii, which form the C. complex, are emerging etiologic agents of fungal infections known to be resistant most commonly used antifungals. The well-established anti-Candida potential metal complexes containing 1,10-phenanthroline (phen) ligands encouraged us evaluate different copper(II), manganese(II), silver(I) phen chelates for their ability inhibit planktonic growth biofilm species complex. Two novel coordination complexes,...
ObjectivesThe beneficial antimicrobial properties of 1,10-phenanthroline (phen)-based drugs, together with the imperative need to develop new chemotherapeutic options for prevention/treatment infections caused by MDR Gram-negative bacteria, led us evaluate effects phen, 1,10-phenanthroline-5,6-dione (phendione), [Ag(phendione)2]ClO4 and [Cu(phendione)3](ClO4)2·4H2O on planktonic- biofilm-growing Pseudomonas aeruginosa.
Reported herein is the ability of Scedosporium apiospermum, S. aurantiacum, minutisporum and Lomentospora prolificans conidia to adhere, differentiate into hyphae form biofilms on both polystyrene lung epithelial cells. To different degrees, all fungi adhered after 4 h, with a predominance those germinated conidia. Prolonged fungi–polystyrene contact resulted in formation monolayer intertwined mycelia, which was identified as typical biofilm structure due presence viable mycelial biomass,...
Elastase B (lasB) is a multifunctional metalloenzyme secreted by the gram-negative pathogen Pseudomonas aeruginosa, and this enzyme orchestrates several physiopathological events during bacteria-host interplays. LasB considered to be potential target for development of an innovative chemotherapeutic approach, especially against multidrug-resistant strains. Recently, our group showed that 1,10-phenanthroline-5,6-dione (phendione), [Ag(phendione)2]ClO4 (Ag-phendione)...
Although considered rare, the emergent Candida haemulonii species complex, formed by C. sensu stricto (Ch), duobushaemulonii (Cd) and var. vulnera (Chv), is highlighted due to its profile of increased resistance available antifungal drugs. In present work, 25 clinical isolates, recovered from human infections during 2011–2020 biochemically identified automated system as haemulonii, were initially assessed molecular methods (amplification sequencing ITS1-5.8S-ITS2 gene) for precise...
Background Leishmania is the etiologic agent of leishmanisais, a protozoan disease whose pathogenic events are not well understood. Current therapy suboptimal due to toxicity available therapeutic agents and emergence drug resistance. Compounding these problems increase in number cases Leishmania-HIV coinfection, overlap between AIDS epidemic leishmaniasis. Methodology/Principal Findings In present report, we have investigated effect HIV aspartyl peptidase inhibitors (PIs) on amazonensis...
In the current study, we evaluated mechanism of action miltefosine, which is first effective and safe oral treatment for visceral leishmaniasis, in Leishmania amazonensis promastigotes. Miltefosine induced a process programmed cell death, was determined by externalization phosphatidylserine, incorporation propidium iodide, cell-cycle arrest at sub-G0/G1 phase DNA fragmentation into oligonucleosome-sized fragments. Despite intrinsic variation that detected spp, our results indicate...
Candida haemulonii is an emerging opportunistic pathogen resistant to most antifungal drugs currently used in clinical arena. Metal complexes containing 1,10-phenanthroline (phen) chelating ligands have well-established anti-Candida activity against different medically relevant species. This study utilized larvae of Galleria mellonella, a widely model vivo infection, examine C. infection characteristics response copper(II), manganese(II) and silver(I) chelates phen, which had demonstrated...
The polyene amphotericin B (AMB) exerts a powerful and broad antifungal activity. AMB acts by (i) binding to ergosterol, leading pore formation at the fungal plasma membrane with subsequent ion leakage, (ii) inducing intracellular accumulation of reactive oxygen species (ROS). Herein, we have deciphered resistance mechanisms in clinical isolates Candida haemulonii complex (C. haemulonii, C. duobushaemulonii, var. vulnera) comparison other clinically relevant non-albicans species. Membrane...
The repurposing strategy was applied herein to evaluate the effects of lopinavir, an aspartic protease inhibitor currently used in treatment HIV-infected individuals, on globally widespread opportunistic human fungal pathogen Candida albicans by using silico, vitro and vivo approaches order decipher its targets cells antifungal mechanisms action. Secreted proteases (Saps) are obviously main target lopinavir. To confirm this hypothesis, molecular docking assays revealed that lopinavir bound...
Leishmaniasis is a neglected disease caused by protozoa belonging to the Leishmania genus. Notably, search for new, promising and potent anti-Leishmania compounds remains major goal due inefficacy of available drugs used nowadays. In present work, we evaluated effects 1,10-phenanthroline-5,6-dione (phendione) coordinated silver(I), [Ag(phendione)2]ClO4 (Ag-phendione), copper(II), [Cu(phendione)3](ClO4)2·4H2O (Cu-phendione), as potential be in chemotherapy against amazonensis chagasi. The...
