To identify the sites in p53 tumor suppressor gene most susceptible to carcinogenic mutation by sunlight, entire coding region of 27 basal cell carcinomas (BCCs) skin was sequenced. Fifty-six percent tumors contained mutations, and these were UV-like: primarily CC-->TT or C-->T changes at dipyrimidine sites. Such mutations can alter more than half 393 amino acids p53, but two-thirds occurred nine which seen once BCC previously studied squamous skin. Seven hotspots specific cancers....

Purpose Circulating cell-free DNA is present in increased amounts the blood of cancer patients, but clinical relevance this phenomenon remains unclear. We conducted a study to assess value circulating as prognostic marker patients with non–small-cell lung (NSCLC). Patients and Methods A standard protocol for quantification by real-time polymerase chain reaction was set up validated at two oncology units. One hundred eighty-five informed NSCLC 46 healthy controls were included study....

Background: The emergence of resistance to chemotherapy remains a major problem in the treatment patients with small-cell lung cancer. Elevated expression Bcl-2, protein that inhibits programmed cell death or apoptosis, has been associated radiation and drug observed majority cancer specimens lines. Purpose: To test hypothesis Bcl-2 levels are critical for inhibiting apoptosis cells, we used an antisense strategy reduce these cells attempt restore natural occurrence apoptosis. Methods:...

Background:Basal cell carcinoma (BCC) of the skin is most common cancer in humans. Epidemiologic studies implicate sunlight exposure as one risk factor, but limited association between BCCs and UVB radiation (i.e., UV a wavelength 280–320 nm) suggests that additional factors must be involved. At molecular level, not much known about role specific environmental agents pathogenesis BCCs. Point mutations types produced by are seen p53 gene (also TP53; chromosome 17p) 40%–56% Loss heterozygosity...

Background: Increased expression of the anti-apoptotic proteins Bcl-2 and Bcl-xL is involved in development progression many tumors. We recently reported that bcl-2/bcl-xL-bispecific antisense oligonucleotide 4625 induces apoptosis lung carcinoma cells. To further assess therapeutic potential 4625, we investigated its effect on a series human tumor cell lines diverse histologic origins vitro vivo. Methods: Oligonucleotide 4625-mediated inhibition bcl-2 bcl-xL was measured breast cells with...

Serum biomarkers can improve diagnosis and treatment of malignant pleural mesothelioma (MPM). However, the evaluation potential new serum biomarker candidates is hampered by a lack assay technologies for their clinical evaluation. Here we followed hypothesis-driven targeted proteomics strategy identification MPM candidate in patient cohorts.Based on hypothesis that cell surface exposed glycoproteins are prone to be released from tumor-cells circulatory system, screened surfaceome model lines...

Expression of the urokinase-type plasminogen activator (uPA) gene in LLC-PK1 cells can be induced by signals mediated both cAMP-dependent protein kinase (PKA) and Ca(2+)- phospholipid-dependent (PKC). We have utilized tumor promoter 12-O-tetradecanoylphorbol 13-acetate (TPA) to down-regulate PKC, order test for an effect on PKA-mediated induction uPA expression. Incubation 24 h with 100 ng/ml TPA caused a marked decrease PKC protein, cytosolic particulate fractions, 85% reduction total...

In the porcine renal epithelial cell line, LLC-PK1, activation of cAMP-dependent signal transduction pathway induces urokinase-type plasminogen activator (uPA) gene. We show here that cAMP response is enhanced when intracellular calcium concentration increased. When LLC-PK1 cells were treated with ionophore ionomycin alone, there was no uPA mRNA accumulation. However, in presence dose-response 8-bromo-cAMP (Br-cAMP) respect to accumulation shifted toward lower concentrations Br-cAMP. A...

Teneurins are a family of highly conserved pair-rule proteins involved in morphogenesis and development the central nervous system. Their function adult tissues disease is largely unknown. Recent evidence suggests role for dysregulated expression human tumors, but systematic investigations missing. Here, we investigated Teneurin-2 Teneurin-4 various cancer cell lines ovarian tumor tissues. were expressed most breast tested. was also detected lines, throughout tumors normal ovary tissue....

Abstract Inorganic arsenic (As) is a toxic xenobiotic and carcinogen associated with severe health conditions. The urban population from the Atacama Desert in northern Chile was exposed to extremely high As levels (up 600 µg/l) drinking water between 1958 1971, leading increased incidence of urinary bladder cancer (BC), skin cancer, kidney coronary thrombosis decades later. Besides, Andean Native-American ancestors were previously for millennia elevated (∼120 at least 5,000 years, suggesting...
