近年,漢方薬は様々な疾患に幅広く用いられ,その中でも五苓散は漢方医学において体内の水分代謝調節作用を持ち,生体恒常性の維持に有意に作用するとされ,臨床的にも使用頻度の高い漢方方剤のひとつである.また,多くの疾患がチャネル遺伝子の異常であることが明らかになり,正常なチャネルの局在が生理機能の恒常性維持に重要であることが分ってきた.そのひとつに腎障害があり,腎臓の集合管に存在する水チャネルであるアクアポリン(aquaporin:AQP)の発現異常により尿濃縮障害が生じることも証明されている.また,慢性腎不全モデルである5/6腎摘ラットにおいて集合管でのアクアポリン2のmRNAおよびタンパク質発現量が比較的保存されていることが報告され,これが残存尿濃縮力を支えていることが示唆されている.そこで本研究では高血糖状態を介さずに,線維化を惹起する5/6腎摘ラットを用い,五苓散の尿濃縮機能と腎組織中のアクアポリン1~3タンパク質発現量,さらに腎障害進展抑制作用に対する五苓散の影響について検討した.五苓散を投与することにより,尿中タンパク量を低下させることから,腎障害進展抑制作用を有する.また,...
