Abstract Premature termination codons (PTCs) prevent translation of a full-length protein and trigger nonsense-mediated mRNA decay (NMD). Nonsense suppression (also termed readthrough) therapy restores function by selectively suppressing at PTCs. Poor efficacy current readthrough agents prompted us to search for better compounds. An NMD-sensitive NanoLuc reporter was used screen 771,345 Among the 180 compounds identified with activity, SRI-37240 its more potent derivative SRI-41315, induce...

Unbalanced apoptosis is a major cause of structural remodelling vasculatures associated with PAH (pulmonary arterial hypertension), whereas the underlying mechanisms are still elusive. miRNAs (microRNAs) regulate expression several proteins that important for cell fate, including differentiation, proliferation and apoptosis. It possible these regulatory RNA molecules play role in development PAH. To test this hypothesis, we studied effect on cultured PASMCs artery smooth muscle cells)...

PGC-1alpha (peroxisome proliferator-activated receptor [PPARgamma] coactivator-1alpha) activates PPARalpha and mitochondrial transcription factor A (Tfam), which regulate proteins, fatty acid ATP metabolism (i.e., FAT/CD36, MCAD, COX I). Recently we found that the salutary effects of estradiol (E2) on cardiac function following trauma-hemorrhage (T-H) are mediated via estrogen (ER)beta. In this study tested hypothesis ERbeta-mediated cardioprotection is induced up-regulation through or...

We found that forskolin-induced swelling in G550X-CFTR patient-derived intestinal organoids (PDOs) was significantly higher than G542X-CFTR PDOs after treatment with ELX-02. Tryptophan (W) the sole amino acid inserted G550X position readthrough. Resulting G550W-CFTR protein exhibited supernormal CFTR activity, PKA sensitivity, and open probability. These results show aminoglycoside-induced readthrough of produces greater function because gain-of-function properties product.

Helicobacter pylori (H. pylori) is one of the most prevalent pathogens globally, and long-term infection causes various gastrointestinal diseases such as gastritis even cancer. In present study, we screened dozens lactic acid bacteria for efficacy to inhibit H. growth in vitro, tested therapeutic effects candidate strains vivo. The results showed that Limosilactobacillus fermentum MN-LF23 (LF23) Lactobacillus gasseri MN-LG80 (LG80) significantly reduced abundance by 90% 83% infected mice,...

Although angiotensin II (Ang II) plays a key role in development of organ ischemia-reperfusion injury, it remains unclear whether is involved intestinal injury following trauma-hemorrhage (T-H). Studies have shown that 17beta-estradiol (E2) administration T-H improves small blood flow; however, Ang this E2-mediated salutary effect. Male Sprague-Dawley rats underwent laparotomy and hemorrhagic shock (removal 60% total volume, fluid resuscitation after 90 min). At onset resuscitation, were...

Although 17beta-estradiol (estrogen) and estrogen receptor (ER) agonist administration after trauma-hemorrhage improves cardiac function, it remains unknown what the optimal or ER dosage is to elicit this beneficial effect. To study this, dose-dependent effects of estrogen, propylpyrazole triol (ER-alpha agonist), diarylpropionitrile (DPN; ER-beta agonist) on heart performance (+dP/dt) were determined in sham rats experimental animals at time maximal bleedout (MBO) 2 h trauma-hemorrhage. Abstract Hepatic damage occurs in males and ovariectomized (OVX), not proestrus (PE), females following trauma-hemorrhage (T-H). The mechanism responsible for hepatoprotection remains unknown. We hypothesized protection PE is a result of enhanced heme oxygenase-1 (HO-1)-derived down-regulation liver inflammatory responses. OVX rats underwent T-H (midline laparotomy, 60% blood loss). received vehicle (Veh; saline), HO-1 inhibitor chromium mesoporphyrin IX chloride (CrMP; 2.5 mg/kg), zinc...
