We have investigated the electrical conductivity of lambda-DNA using DNA covalently bonded to Au electrodes. Thiol-modified dTTP was incorporated into `sticky' ends bacteriophage polymerase. Two-probe measurements on such molecules provide a hard lower bound for resistivity \rho>10^6 (Ohm cm) at bias potentials up 20 volts, in conflict with recent claims moderate high conductivity. By direct imaging, we show that are present after measurements. stress importance eliminating salt residues these
The effect of confinement on the dynamics polymers was studied by observing transient extension and relaxation single DNA molecules as they interacted with obstacles in a specially designed thin slit. Viscous drag found to increase degree confinement, which we interpret terms hydrodynamic screening planar surfaces Since driven an electrophoretic force, experimental data support notion that electric field acts tethered polyelectrolyte equivalently flow.

X-ray absorption spectroscopy at the Zn K-edge indicates that active site of marine diatom Thalassiosira weissflogii carbonic anhydrase is strikingly similar to mammalian α-carbonic enzymes. The zinc has three histidine ligands and a single water 1.98 Å. This quite different from β-carbonic anhydrases higher plants in which coordinated by two cysteine thiolates, one histidine, molecule. shows no significant sequence similarity with other may represent an example convergent evolution molecular level.
Randomly distributed Dictyostelium discoideum cells form cooperative territories by signaling to each other with cAMP. Cells initiate the process sending out pulsatile signals, which propagate as waves. With time, circular and spiral patterns form. We show that adding spatial temporal noise levels of an important regulator external cAMP levels, phosphodiesterase inhibitor, we can explain natural progression system from randomly firing waves whose symmetries break double- single- or...
The thermodynamics of Zn2+ binding to three peptides corresponding naturally occurring Zn-binding sequences in transcription factors have been quantified with isothermal titration calorimetry (ITC). These peptides, the third zinc finger Sp1 (Sp1-3), second myelin factor 1 (MyT1-2), and sequence DNA-binding domain glucocorticoid receptor (GR-2), bind Cys2His2, Cys2HisCys, Cys4 coordination, respectively. Circular dichroism confirms that Sp1-3 MyT1-2 considerable negligible Zn-stabilized...
Recent data on the physiology of inorganic carbon acquisition by model marine diatom Thalassiosira weissflogii (Grunow) demonstrate importance catalytic equilibration HCO3-and CO2by carbonic anhydrases located in periplasm and cytoplasm. These enzymes can use Zn, Co or Cd as their metal centre, activity increases at low ambient CO2. The silica frustule provides buffering for extracellular CA activity, transmembrane transport CO2 may occur passive diffusion. Under limitation, cytoplasmic...
ABSTRACT In this paper we report on the isolation and genetic analysis of a series strong mutators mapping at five minutes E. coli chromosome. These mutations are dominant show no evidence interaction in merodiploids. Cultures grown broth medium exhibit mutant frequencies to six orders magnitude higher than mut+ strains. propagated minimal salts media mutate rates one three wild-type. Three-factor crosses have been used order these relative metD, proA, Tn10 insertion near minutes.
This paper reports on the separation of Dictyostelium discoideum chromosomes by pulse-field electrophoresis and correlation electrophoretic pattern with linkage groups established classical genetic methods. In two commonly used laboratory strains, five chromosome-sized DNA molecules have been identified. Although majority molecular probes in this study can be unambiguously assigned to groups, karyotype differs between closely related strains AX3k NC4, suggesting that chromosomal...

Micro- and nanofabrication techniques have provided an unprecedented opportunity to create a designed world in which separation fractionation technologies normally occur on the macroscopic scale can be optimized by designing structures utilize basic physics of process, or new processes realized building do not exist without external design. Since microfabrication is exceedingly sophisticated its development, it possible design construct highly creative microdevices allow one probe specific...
We outline some of the possible applications nanotechnology to modern molecular biology and discuss several technologies that can be used make nanoscale confining environments (channels or post arrays) for long polymers such as DNA. A particular emphasis is placed on making large arrays using non-electron beam lithography methods. then how focused ion (FIB) milling construct nearfield slits examining molecules.
Animal Computer Interaction (ACI) aims to design user-centered interactions between animals and technology. In this regard, a major challenge for researchers is accurately assessing interpreting animal behavior, in part, due the invasive nature of data collection techniques individuality behavior. This paper presents method that uses tail wagging, communicative behavior dogs used welfare studies, as non-invasive parameter measure canine user experience (UX). We present findings from study...
The region of the Escherichia coli chromosome coding for mutD gene was cloned, mutD5 function resides on a 1.2-kilobase fragment 28-kilodalton (kDa) protein. A deletion end-point analysis shows that presence 28-kDa protein is required and suggests functional has sufficient capacity to code second polypeptide approximately 20 kDa. Plasmids carrying mut+ alleles both produce species. product dominant when carried by single multicopy plasmids. only Thus, exhibits negative complementation. We...
Abstract The sedimentation coefficient of T2 phage DNA has been studied by zone centrifugation in sector‐shaped preparative centrifuge tubes over a concentration range 0.02–2.0 μg./ml. DNA. These results have compared to similar study the analytical T4 0.50–5.75 A value for 60.7 ± 1.8 S. was obtained first method and 61.3 1.5 second.
We have observed electrostatic trapping of tribranched DNA molecules undergoing electrophoresis in a microfabricated pseudo-two-dimensional array posts. Trapping occurs unique transport regimen which the electrophoretic mobility is extremely sensitive to polymer topology. The arrest branched polymers explained by considering their center-of-mass motion; certain conformations, owing constraints imposed obstacles molecule cannot advance without center mass first moving short distance...
Complementation tests were performed between pairs of independently isolated aggregation defective mutants P. violaceum. Analysis the results suggests that approximate number genes controlling in this organism is 50.
Dogs are being increasingly exposed to interactive technology aiming entertain them with playful experiences. However, work that investigates how dogs engage tangible play interfaces, whether they manifest different interaction patterns and these might relate their personality has so far been limited. In this paper, we explore interact a novel physical game. Our study involved 60 participants, whose was assessed using standard test interactions behaviours during the game were observed....
