Metazoan E(y)2/ENY2 is a multifunctional protein important for transcription activation and mRNA export, being component of SAGA/TFTC the export complex AMEX. Here, we show that ENY2 in Drosophila also stably associated with THO, involved mRNP biogenesis. The ENY2–THO required normal development, functioning independently on SAGA THO arrive transcribed region hsp70 gene after its activation, plays an role recruitment. no direct association elongating RNA polymerase II. Recruitment occurs by...

Despite increasing data on the properties of replication origins, molecular mechanisms underlying origin recognition complex (ORC) positioning in genome are still poorly understood. The Su(Hw) protein accounts for activity best-studied Drosophila insulators. Here, we show that recruits histone acetyltransferase SAGA and chromatin remodeler Brahma to Su(Hw)-dependent insulators, which gives rise regions with low nucleosome density creates conditions ORC binding. Depletion leads a dramatic...

The origin recognition complex (ORC) of eukaryotes associates with the replication origins and initiates pre-replication assembly. In literature, there are several reports interaction ORC different RNAs. Here, we demonstrate for first time a direct THSC/TREX-2 mRNA nuclear export complex. was purified from Drosophila embryonic extract found to bind fraction ORC. This occurred via subunits essential viability. Also, associated mRNP, which facilitated by TREX-2. interacted Nxf1 receptor...

AbstractThe Drosophila TATA box-binding protein (TBP)-related factor 2 (TRF2 or TLF) was shown to control a subset of genes different from that controlled by TBP. Here, we have investigated the structure and functions trf2 gene. We demonstrate it encodes two isoforms: previously described 75-kDa TRF2 newly identified 175-kDa version in which same sequence is preceded long N-terminal domain with coiled-coil motifs. Chromatography embryo extracts revealed part multiprotein complex also...

SAGA/TFTC is a histone acetyltransferase complex that has second enzymatic activity because of the presence deubiquitination module (DUBm). Drosophila DUBm consists Sgf11, ENY2 and Nonstop proteins. We show Sgf11 other DUBm-independent functions. It associates with Cbp80 component cap-binding thereby recruited onto growing messenger ribonucleic acid (mRNA); it also interacts AMEX mRNA export essential for hsp70 export, as well general from nucleus. Thus, functions both SAGA biogenesis machinery.

The germline-specific role of telomeres consists chromosome end elongation and proper segregation during early developmental stages. Despite the crucial in germ cells, little is known about telomere biology germline. We analyzed homeostasis Drosophila female germline embryos. A novel function deadenylase complex Ccr4-Not telomeric transcript surveillance mechanism reported. Depletion components causes strong derepression retroelement HeT-A accompanied by poly(A) tail. ENY2/Sus1, a protein involved in the coupling of transcription with mRNA export, is component SAGA/TFTC and TREX-2/AMEX complexes. Recently, we have described association ENY2 third complex, THO. Moreover, our data indicate that also associated other factors, both nucleus cytoplasm. Thus, being shared components several complexes, appears to function as an adapter molecule integration cellular processes, particular, subsequent stages gene expression.

The different steps of gene expression are intimately linked to coordinate and regulate this complex process. During transcription, numerous RNA-binding proteins already loaded onto the nascent mRNA package into a messenger ribonucleoprotein particle (mRNP). These often also involved in other than packaging. For example, TREX functions mRNP packaging nuclear export. Previously, we showed that Prp19 splicing (Prp19C) is needed for efficient transcription as well occupancy at transcribed...
