All sequenced histidine tRNAs have one additional nucleotide at the 5' end compared with other tRNA species. To investigate role of this unique structure in aminoacylation, we constructed vitro transcripts corresponding to E. coli sequence and its variants G-1-C73 base pair, by using T7 RNA polymerase transcription system. A transcript having a wild-type no modified bases was good substrate for histidyl-tRNA synthetase (HisRS), aminoacylation activity affected introduction triphosphate...

The stable RNAs, whose sequences are homologous to 10Sa RNA of Escherichia coli , have been isolated from Mycoplasma capricolum and Bacillus subtilis both belonging the Gram-positive bacterial group. total nucleotide RNAs determined by partial sequencing DNA their genes. A comparison sequences, together with those other so far known, has shown that 5′- 3′-end well conserved among species, while central parts reveal little homologies. Unexpectedly, 3′-regions can be folded in a common...

Crocin, picrocrocin, and safranal as the major secondary metabolites are important for high quality of spice saffron. Crocin picrocrocin were detected in stigma-like structures proliferated vitro. Safranal appeared after heat treatment. The contents relative ratio these substances similar to those intact young stigmas, indicating that showed not only morphological but also biochemical similarity stigmas.

Ribosome biogenesis is a tightly regulated, multi-stepped process. The assembly of ribosomal subunits central step the complex process, involving nearly 30 protein factors in vivo bacteria. Although process has been extensively studied vitro for over 40 years, very limited information known and specific roles factors. Such an example ribosome maturation factor M (RimM), involved late-stage 30S subunit. Here, we combined quantitative mass spectrometry cryo-electron microscopy to characterize...

Bacterial transfer-messenger RNA (tmRNA, 10Sa RNA) is involved in a trans-translation reaction which contributes to the degradation of incompletely synthesized peptides and recycling stalled ribosomes. However, its physiological role cell remains elusive. In this study, an efficient system for controlling expression gene tmRNA (ssrA), as well gene-defective strain (ssrA:cat), were constructed Bacillus subtilis. The effects on growth cells investigated under various culture conditions using...

Abstract The 16,260-bp mitochondrial DNA (mtDNA) from the starfish Asterina pectinifera has been sequenced. genes for 13 proteins, two rRNAs and 22 tRNAs are organized in an extremely economical fashion, similar to those of other animal mtDNAs, with some overlapping each other. gene organization is same as that another echinoderm, sea urchin, except inversion a 4.6-kb segment contains 16S rRNA. Judging protein coding genes, mammalian mtDNAs resemble urchin mtDNA more than starfish. region...

A novel translation, trans ‐translation, is facilitated by a highly structured RNA molecule, tmRNA. This molecule has two structural domains, tRNA domain and an mRNA domain, the latter including four pseudoknot structures (PK1 to PK4). Here, we show that replacement of each these pseudoknots, except PK1, in Escherichia coli tmRNA with single stranded did not seriously affect functions as alanine mRNA. Furthermore, three pseudoknots were interchangeable only small losses functions. These...

Recognition of tRNA His by Saccharomyces corevisiae histidyl-tRNA synthetase was studied using in vitro transcripts. Histidine is unique possessing an extra nucleotide, G 1 , at the 5′ end. Mutation studies indicate that this irregular secondary structure end acceptor stem important for aminoacylation with histidine, while requirement either base pair smaiier than Escherichia coli . The anticodon also found to be required histidylation. regions involved histidyiation are essentially same as...

Although trans-translation mediated by tmRNA-SmpB has long been known as the sole system to relieve bacterial stalled ribosomes, ArfA recently identified an alternative factor for ribosome rescue in Escherichia coli. This process requires hydrolysis of nascent peptidyl-tRNA RF2, which usually acts a stop codon-specific peptide release factor. It poses fascinating question how and RF2 recognize ribosome. Here, we mapped location directed hydroxyl radical probing. revealed ArfA-binding site...
