ABSTRACT This study evaluated cross-resistance of Mycobacterium tuberculosis strains to ofloxacin, moxifloxacin, and gatifloxacin investigated the presence mutations in gyrA gyrB . Fluoroquinolone susceptibilities were determined for 41 M. by proportion method on 7H11, MICs resazurin microtiter assay. Forty shared same resistance results three fluoroquinolones. However, one strain, with an Asn-533 → Thr mutation , was susceptible ofloxacin but resistant moxifloxacin gatifloxacin.
ABSTRACT The MIC for streptomycin in the presence of efflux pump (EP) inhibitors and sequencing rpsL , rrs gidB genes provided evidence possible participation EP low-level (STR) resistance some isolates without mutations. Mutation gene an could act synergistically to confer low STR resistance.
Abstract Background Mutations associated with resistance to rifampin or streptomycin have been reported for W/Beijing and Latin American Mediterranean (LAM) strain families of Mycobacterium tuberculosis . A few studies limited sample sizes separately evaluated mutations in kat G, ahp C inh genes that are isoniazid (INH) resistance. Increasing prevalence INH resistance, especially high (TB) prevalent countries is worsening the burden TB control programs, since similar transmission rates noted...
Helicobacter pylori-induced gastric mucosal inflammation is mediated by proinflammatory and anti-inflammatory cytokines. Polymorphisms in genes that code cytokines influence cytokine secretion levels appear to contribute the risk of diseases. In this sense, we performed study identify polymorphisms IL-6, IL-8, IL-10 their associations with H. pylori infection pathologies.Gastric biopsy samples 151 patients infected 76 uninfected individuals were used. was diagnosed histological examination...
The municipality of Oriximiná, Brazil, has 33 quilombola communities in remote areas, endowed with wide experience the use medicinal plants. An ethnobotanical survey was carried out five these communities. A free-listing method directed for species locally indicated against Tuberculosis and lung problems also applied. Data were analyzed by quantitative techniques: saliency index major agreement. Thirty four informants related 254 ethnospecies. Among these, 43 surveyed possible...
Twenty-three naphthoimidazoles and six naphthoxazoles were synthesised evaluated against susceptible rifampicin- isoniazid-resistant strains of Mycobacterium tuberculosis. Among all the compounds evaluated, fourteen presented MIC values in range 0.78 to 6.25 μg/mL resistant M. Five structures solved by X-ray crystallographic analysis. These substances are promising antimycobacterial prototypes.
Helicobacter pylori infects nearly 50% of the world's population. This microorganism is accepted as most important agent gastritis and a risk factor for peptic ulcer disease gastric adenocarcinoma. Currently many diagnostic methods exist detecting H. pylori, however they all have limitations, thus it recommend combination at least two methods. The aim this study was to evaluate methods, such in-house urease test, culture Polymerase Chain Reaction (PCR), detection in biopsy specimens 144...
The basis of drug resistance in Mycobacterium abscessus is still poorly understood. Nevertheless, as seen other microorganisms, the efflux antimicrobials may also play a role M. resistance. Here, we investigated pumps clarithromycin using nine clinical isolates complex belonging to T28 erm(41) sequevar responsible for inducible clarithromycin. strains were characterized by susceptibility testing presence/absence inhibitor verapamil and genetic analysis drug-resistance-associated genes....
Duroia macrophylla popularly known as “cabeça-de-urubú,” “apuruí,” or “puruí-grande-da-mata” occurs in the Amazon Forest. Its leaves and branches were collected twice extracted with dichloromethane methanol. All extracts subjected to phytochemical investigation terpenes flavonoids found all methanol extracts, respectively. Methanol from both (1st collection) (2nd presented hydrolyzed tannins, yet alkaloids only detected at 2nd collection. Phenol compounds extracts’ collections. The action of...
ABSTRACT Spoligotyping has shown Mycobacterium tuberculosis strains to be composed of different lineages, and some them are not just geographically restricted but also affect specific ethnic populations associated with outbreaks drug resistance. We recently described a particular subtype within the Latin American-Mediterranean (LAM) family, called RD Rio , widespread in Brazil. Moreover, recent data indicate that is present many countries on all continents cavitary disease multidrug...
Forty eight ethanolic crude extracts and fractions (hexane, dichloromethane, ethyl acetate n-butanol) from ten Brazilian plants (Leguminosae, Monimiaceae Verbenaceae), 1 Costa Rica (Verbenaceae) Argentina were screened for anti-mycobacterium activity against Mycobacterium tuberculosis (ATCC-27294H37Rv), by the Alamar Blue test, at a fixed concentration of 100 µg/mL. Out forty eight, seven active this concentration, corresponding to Lantana trifolia (hexane dichloromethane leaves), Vitex...
Tuberculosis remains a serious public health problem, with nine million cases being reported annually. Treatment antibiotics is the most effective mechanism to control this disease, although increase in resistant strains, co-infection HIV, and long duration of treatment has established need develop new drugs. Here we show activity usnic acid against susceptible Mycobacterium tuberculosis strains nontuberculous mycobacteria. Further, did not identify any contribution efflux innate resistance acid.
We standardized a method to evaluate the growth kinetics of Mycobacterium tuberculosis by measuring quantitatively reduction resazurin spectrophotometry. Growth curves and rate twenty-one M. clinical isolates were determined. The showed technical simplicity is inexpensive assess fitness each isolate.
Background: Tuberculosis is a worldwide public health problem and more prevalent in specific populations, such as prisoners. The aim of this study was to analyze the epidemiological operational indicators tuberculosis prisoners southern region Brazil. Methods: This descriptive, observational study, utilizing secondary data from Notifiable Diseases Information System on cases diagnosed state Rio Grande do Sul, Brazil, 2014 2018. Prisoner used calculate incidence were extracted reports by...
Thirty-six plant extracts from the brazilian Atlantic Forest were tested for their antimycobacterial activity against Mycobacterium tuberculosis H37Rv and M. kansasii, using method REMA in seriate concentrations of 100 to 0.20 µg/mL. Among thirty six tested, five active tuberculosis, three these also showed kansasii. Cytotoxicity test with VERO cells was performed tuberculosis. Only extract Peschiera affinis identified as non-toxic concentration 100µg/mL.
RDRio is a novel Mycobacterium tuberculosis lineage of the Latin American–Mediterranean (LAM) family. LAM has been found worldwide but more predominant in South America. The aim this study was to assess presence and family city Rio Grande, Brazil, investigate fitness these strains based on determination their growth rate. Fifty clinical isolates M. were genotyped 43 different patterns by spoligotyping mycobacterial interspersed repetitive units–variable number tandem repeats. genotypes...
