Abstract Background The Direct Repeat locus of the Mycobacterium tuberculosis complex (MTC) is a member CRISPR (Clustered regularly interspaced short palindromic repeats) sequences family. Spoligotyping widely used PCR-based reverse-hybridization blotting technique that assays genetic diversity this and useful both for clinical laboratory, molecular epidemiology, evolutionary population genetics. It easy, robust, cheap, produces highly diverse portable numerical results, as result...
ABSTRACT The present update on the global distribution of Mycobacterium tuberculosis complex spoligotypes provides both octal and binary descriptions for M. complex, including bovis , from >90 countries (13,008 patterns grouped into 813 shared types containing 11,708 isolates 1,300 orphan patterns). A number potential indices were developed to summarize information biogeographical specificity a given type, as well its geographical spreading (matching code index, respectively). To...
Abstract We present a short summary of recent observations on the global distribution major clades Mycobacterium tuberculosis complex, causative agent tuberculosis. This was defined by data-mining an international spoligotyping database, SpolDB3. database contains 11,708 patterns from as many clinical isolates originating more than 90 countries. The spoligotypes were clustered into 813 shared types. A total 1,300 orphan (clinical showing unique spoligotype) also detected.
ABSTRACT The MIC for streptomycin in the presence of efflux pump (EP) inhibitors and sequencing rpsL , rrs gidB genes provided evidence possible participation EP low-level (STR) resistance some isolates without mutations. Mutation gene an could act synergistically to confer low STR resistance.
Drug resistant tuberculosis continues to increase and new approaches for its treatment are necessary. The identification of M. clinical isolates presenting efflux as part their phenotype has a major impact in treatment. In this work, we used checkerboard procedure combined with the tetrazolium microplate-based assay (TEMA) study single combinations between antituberculosis drugs inhibitors (EIs) against multidrug using fully susceptible strain H37Rv reference. Efflux activity was studied on...
Numerous studies show efflux as a universal bacterial mechanism contributing to antibiotic resistance and also that the activity of antibiotics subject can be enhanced by combined use inhibitors. Nevertheless, contribution overall drug levels clinical isolates Mycobacterium tuberculosis is poorly understood still ignored many. Here, we evaluated plus target-gene mutations in M. tuberculosis. A panel 17 strains were characterized for associated profiles presence absence The correlation...
ABSTRACT The presence of mutations in specific regions the katG , inhA and ahpC genes was analyzed with 69 Mycobacterium tuberculosis isoniazid-resistant isolates from three Brazilian states. Point codon 315 gene were observed 87.1, 60.9, 60% Rio Grande do Sul, de Janeiro, São Paulo, respectively. Mutations identified only one isolate RJ State, promoter region revealed distinct positions 12.9, 21.7, 6.7% RS, SP,
ABSTRACT Mutations related to streptomycin resistance in the rpsL and rrs genes are well known can explain about 70% of this phenotypic resistance. Recently, gidB gene was found be associated with low-level Mycobacterium tuberculosis . have been reported high frequency, appears very polymorphic, frameshift point mutations occurring streptomycin-susceptible streptomycin-resistant strains. In study, appeared 27% strains that contained no or genes, they were However, association certain needs...
Abstract Background Developments in molecular detection and strain differentiation of members Mycobacterium tuberculosis complex have proved to be useful. The DNA extraction method influences the amplification efficiency, causing interference on sensitivity respective inhibitors. aim this study was standardize a simple fast method, providing by IS6110-PCR effectively free from undue interferences. Findings efficiency six different protocols tested M. cultures has varied 75% 92.5%. This...
The frequency of the Beijing genotype Mycobacterium tuberculosis as a cause (TB) in South America was determined by analyzing genotypes strains isolated from patients that had been diagnosed with disease between 1997 and 2003 seven countries subcontinent. In total, 19 1,202 (1.6%) TB cases carried isolates, including 11 185 Peru (5.9%), five 512 Argentina (1.0%), two 252 Brazilian (0.8%), one 166 Paraguay (0.6%) none samples obtained Chile (35), Colombia (36) Ecuador (16). Except for were...
In Mycobacterium tuberculosis (MTB) infection, the complex interaction of host immune system and mycobacteria is associated with levels cytokines production that play a major role in determining outcome disease. Several single-nucleotide polymorphisms (SNPs) cytokine genes have been (TB) outcome. The aim this study was to evaluate association between previously reported SNPs IL2–330 T>G (rs2069762); IL4–590 C>T (rs2243250); IL6–174 G>C (rs1800795); IL10–592 A>C (rs1800872); IL10–1082 G>A...
The influence of different infectious agents and their association with human papillomavirus (HPV) in cervical carcinogenesis have not been completely elucidated. This study describes the between cytological changes epithelium detection most relevant aetiological sexually transmitted diseases. Samples collected from 169 patients were evaluated by conventional cytology followed molecular analysis to detect HPV DNA, Chlamydia trachomatis, herpes simplex virus 1 2,Neisseria gonorrhoeae,...
ABSTRACT Mutations in a 69-bp region of the rpoB gene associated with rifampin resistance (Rif r ) 100 isolates (82 Rif from three states Brazil were studied. Twenty-one different kinds mutations identified isolates, and six new alleles are described.
The occurrence of tuberculosis (TB) in prisons has been described as an alarming public health problem many countries, especially developing nations. objective this study was to conduct a survey among prisoners with TB respiratory symptoms order estimate the incidence disease, analyze drug susceptibility profile and genotype isolates Mycobacterium city Charqueadas, southern Brazil. 55/1,900 inhabitants prison; corresponds 3,789/100,000 inhabitants, prevalence 72/1,900 (4,960/100,000...
ABSTRACT Spoligotyping has shown Mycobacterium tuberculosis strains to be composed of different lineages, and some them are not just geographically restricted but also affect specific ethnic populations associated with outbreaks drug resistance. We recently described a particular subtype within the Latin American-Mediterranean (LAM) family, called RD Rio , widespread in Brazil. Moreover, recent data indicate that is present many countries on all continents cavitary disease multidrug...
Although serological assays have been widely used for the diagnosis of canine visceral leishmaniasis (CVL), they present different performances depending on clinical profile dogs. This study evaluated accuracy tests, immunochromatographic (Dual Path Platform: DPP®) and enzyme-linked immunosorbent (ELISA EIE®), CVL in relation to detection Leishmania DNA through real-time polymerase chain reaction (real-time PCR) samples from symptomatic asymptomatic dogs a non-endemic area state Rio Grande...
