MCL-1 (myeloid cell leukemia-1) is an antiapoptotic BCL-2 family protein discovered as early induction gene during myeloblastic leukemia differentiation. This survival has the homology (BH) domains 1, 2, and 3 a C-terminal transmembrane region. We identified short splicing variant of mRNA in human placenta encoding protein, termed (MCL-1S), with altered C terminus compared full-length long (MCL-1L), leading to loss BH1, BH2, domains. Analysis indicated that MCL-1S results from out exon 2...

Abstract Premature ovarian failure in a subgroup of women with blepharophimosis-ptosis-epicanthus inversus type 1 syndrome has been associated nonsense mutations the gene encoding Forkhead transcription factor, L2 (FOXL2). However, exact function FOXL2 ovary is unclear. We investigated expression mouse during follicular development and maturation by RT-PCR situ hybridization. The mRNA expressed ovaries throughout adulthood localized to undifferentiated granulosa cells small medium follicles...

Fluorescence-based amine-reactive dyes are highly valuable for the sensing of amines and labeling biomolecules. Although it would be desirable, large changes in emission spectra intensity seldom accompany conjugation known to their target molecules. On contrary, amide bond formation between pentafluorophenyl (2-PFP) succinimidyl (2-NHS) esters meso-carboxyBODIPY results significant maxima (Δλ: 70–100 nm) (up 3000-fold), enabling fast (down 5 min) selective fluorogenic detection amines, amino...

Malignant melanoma, characterized by its ability to metastasize other organs, is responsible for 90% of skin cancer mortality. To investigate alterations in the cellular metabolome and lipidome related melanoma metastasis, gas chromatography-mass spectrometry (GC-MS) direct infusion-mass (DI-MS)-based metabolic lipidomic profiling were performed on extracts normal human melanocyte (HEMn-LP), low metastatic (A375, G361), highly (A2058, SK-MEL-28) cell lines. Human thyrotropin (TSH), luteotropin (LH), follitropin (FSH), and chorionic gonadotropin are members of the heterodimeric glycoprotein hormone family. The common α subunit forms noncovalent heterodimers with different β subunits. Two novel human hormonelike genes, α2 (A2) β5 (B5), recently have been identified. Using a yeast two-hybrid assay, two subunits were found as potential heterodimerization partners. Immunological analyses confirmed A2 B5 in transfected cells their colocalization...

Mutations in FOXL2 are responsible for blepharophimosis-ptosis-epicanthus inversus syndrome (BPES) type I, which affected women exhibit premature ovarian failure. FOXL2-null mice showed defects granulosa cell development during folliculogenesis. We screened a rat yeast two-hybrid cDNA library to identify FOXL2-interacting proteins and found steroidogenic factor-1 (SF-1). Here, we show that human SF-1 interact cells negatively regulates the transcriptional activation of enzyme, CYP17, by...

The specific detection of eosinophil peroxidase (EPO) activity requires the difficult distinction between hypobromous acid generated by EPO and hypochlorous other haloperoxidases. Here we report a fluorogenic probe that is halogenated with high kinetic selectivity (≥1200:1) for HOBr over HOCl. Heavy-atom effects do not quench dibrominated product because its self-assembly into emissive J-aggregates provide turn-on signal. Applications this fluorogen to assays, dipstick sensors, fluorescence...

A fluorescent probe based on an excimer-forming benzothiazolyl-cyanovinylene (CV) dye was developed to target the apoptotic protease caspase-3. Upon action of caspase-3, water-soluble Ac-DEVD-NH-CV, which is weakly green emissive in aqueous solution, converted hydrophobic CV-NH2, spontaneously aggregates. Aggregation CV-NH2 promotes excimer emission CV dye, allows for study caspase-3 activity vitro and imaging enzyme living cells because large red shift enhanced fluorescence signal probe.
