Succinic acid was produced by fermentation of Anaerobiospirillum succiniciproducens using glycerol as a carbon source. When cells were anaerobically cultured in medium containing 6.5 g/L glycerol, high succinic yield (133%) obtained while avoiding the formation by-product acetic acid. The gram ratio to 25.8:1, which is times higher than that glucose (ca. 4:1) Therefore, can be with much less source, will facilitate its purification. and cofermented increasing decreased, suggesting enhanced...
Abstract Background Phenylpropanoids are the precursors to a range of important plant metabolites such as cell wall constituent lignin and secondary belonging flavonoid/stilbene class compounds. The latter natural products has been shown function in wide biological activities. During last few years an increasing number health benefits have associated with these In particular, they demonstrate potent antioxidant activity ability selectively inhibit certain tyrosine kinases. Biosynthesis many...
Synopsis: A organic carbonate derived from glycerol, ethanol, and CO 2 demonstrates efficient plasticizing properties for PLA, offering a notable advantage in terms of morphological stability over the conventional plasticizer, acetyltributylcitrate.
Abstract Flavonoids are important plant‐specific secondary metabolites synthesized from 4‐coumaroyl coenzyme A (CoA), derived the general phenylpropanoid pathway, and three malonyl‐CoAs. The synthesis involves a plant type III polyketide synthase, chalcone synthase. We report cloning coexpression in Escherichia coli of phenylalanine ammonia lyase, cinnamate‐4‐hydroxylase, 4‐coumarate:CoA ligase, synthase model Arabidopsis thaliana . Simultaneous expression all four genes resulted blockage...
We investigated the cytotoxic activities of crocin and crocetin, 2 major carotenoids isolated from stigma Crocus sativus (saffron), on 5 human cancer cell lines proposed their possible anticancer mechanisms. Crocetin, a glycosylated carotenoid, showed approximately 5- to 18-fold higher cytotoxicity than crocin, carboxylic carotenoid (IC50 0.16-0.61 mmol/L for crocetin vs. 2.0-5.5 crocin). This suggests that structural differences account different efficacies between them....
Steviol glycosides such as stevioside have attracted the attention of food and beverage industry. Recently, efforts were made to produce these natural sweeteners in microorganisms using metabolic engineering. Nonetheless, steviol titer is relatively low metabolically engineered microorganisms, therefore a steviol-biosynthetic pathway heterologous needs be optimized. The purpose this study was redesign reconstruct via synthetic-biology approaches order overproduce Escherichia coli.A...
Abstract Background Crocin, a glycosylated apocarotenoid pigment predominantly found in saffron, has garnered significant interest the field of biotechnology for its bioactive properties. Traditional production crocins and their aglycone, crocetin, typically involves extraction from crocin-producing plants. This study aimed to develop an alternative biosynthetic method these compounds by engineering metabolic pathways zeaxanthin, crocin Escherichia coli strains. Results Employing series...
The biosynthetic pathway for staphyloxanthin, a C30 carotenoid biosynthesized by Staphylococcus aureus, has previously been proposed to consist of five enzymes (CrtO, CrtP, CrtQ, CrtM, and CrtN). Here, we report missing sixth enzyme, 4,4′-diaponeurosporen-aldehyde dehydrogenase (AldH), in the staphyloxanthin describe functional expression complete Escherichia coli. When expressed known through artificial synthetic operons wild-type operon (crtOPQMN) E. coli, aldehyde intermediates such as...
Abstract Recent research interest in phytochemicals has consistently driven the efforts metabolic engineering field toward microbial production of various carotenoids. In spite systematic studies, possibility using C 30 carotenoids as biologically functional compounds not been explored thus far. Here, we generated 13 novel structures and one 35 carotenoid, including acyclic, monocyclic bicyclic structures, through directed evolution combinatorial biosynthesis, Escherichia coli . Measurement...
Promoters regulate the expression of metabolic pathway genes to control flux metabolism. Therefore, fine-tuning gene requires an applicable promoter system. In this study, a dissolved oxygen-dependent nar was engineered for levels biosynthetic enzymes in Escherichia coli. To demonstrate feasibility using synthetic promoters production biochemicals E. coli, d-lactate consisting one enzyme and 2,3-butanediol (BDO) three were investigated. The spacer sequence 15 bp between - 35 10 elements...
Lactic acid has been approved by the United States Food and Drug Administration as Generally Regarded As Safe (GRAS) is commonly used in cosmetics, pharmaceutical, food industries. Applications of lactic have also emerged plastics industry. bacteria (LAB), such Leuconostoc Lactobacillus, are widely producers for food-related biotechnological applications. Nonetheless, industrial mass production LAB a challenge mainly because growth inhibition caused end product, acid. Thus, it important to...
Isoprenoids, which are natural compounds with diverse structures, possess several biological activities that beneficial to humans. A major consideration in isoprenoid production microbial hosts is the accumulation of biosynthesized within intracellular membranes may impede balanced cell growth, consequently reduce desired yield target isoprenoid. As a strategy overcome this suggested limitation, we selected peroxisome as depots for additional storage isoprenoids facilitate increased...
The efficient identification of microbial strains capable producing rare sphingoid bases, such as sphingosine and sphinganine, is critical for advancing fermentation processes addressing increasing industrial demands. Wickerhamomyces ciferrii, a non-conventional yeast, naturally overproduces tetraacetyl phytosphingosine (TAPS); however, the production other valuable including sphingosine, triacetyl remains key target. In this study, we developed novel screening method utilizing fluorescein...
