Intestinal barrier function is important for preserving health, as a compromised allows antigen entry and can induce inflammatory diseases. Probiotic bacteria play role in enhancing intestinal function; however, the mechanisms are not fully understood. Existing studies have focused on ability of probiotics to prevent alterations tight junctions disease models, been restricted few junction bridging proteins. No previously investigated effect probiotic healthy epithelial cell genes involved...

The aim of this research was to identify bacterial isolates having the potential improve intestinal barrier function. Lactobacillus plantarum strains and human oral were screened for their ability enhance tight junction integrity as measured by transepithelial electrical resistance (TEER) assay. Eight commercially used probiotics compared determine which had greatest positive effect on TEER, best-performing probiotic strain, rhamnosus HN001, a benchmark evaluate isolates. One isolate, L. DSM...

Faecalibacterium prausnitzii, an abundant member of the human commensal microbiota, has been proposed to have a protective role in intestine. However, it is obligate anaerobe, difficult co-culture viable form with oxygen-requiring intestinal cells. To overcome this limitation, unique apical anaerobic model barrier, which enabled live anaerobes cell line Caco-2, was developed. Caco-2 cells remained and maintained intact barrier for at least 12 h, consistent gene expression data, suggested had...

Abstract The interplay between diet and fecal microbiota composition is garnering increased interest across various host species, including domestic dogs. While the influence of dietary macronutrients their associated microbial communities have been extensively reviewed, these reviews are descriptive do not account for differences in community analysis, nor they standardize macronutrient content studies. To address this, a meta‐analysis was performed to assess impact crude protein...

Three ovine-derived cathelicidins, SMAP29, OaBac5mini, and OaBac7.5mini, were compared with respect to their antibacterial activities interactions membranes. SMAP29 was confirmed be alpha-helical, broad spectrum, able disrupt both the outer cytoplasmic membranes at relatively low concentrations. In contrast, two proline- arginine-rich OaBac peptides had more-modest activities, reduced levels of lipopolysaccharide binding, a lesser ability depolarize membrane, consistent target.

This study investigates the influence of tomato and feijoa juices as fermentable carriers Lactobacillus plantarum (LP DSM20205) on ability bacterium to improve intestinal barrier function using trans-epithelial electrical resistance (TEER) assay in an apical anaerobic model. The survival LP DSM20205 different fruit during vitro digestion, its adhesion capacity, potential cytotoxic effect Caco-2 cells were also studied. results showed that carrier have a significant growth, TEER. Brain signalling pathways involved in subclinical anxiety and depressed mood can be modulated via the gut brain axis (GBA), providing potential for diet dietary components to affect mood. We investigated behavioural, physiological microbiome responses Lacticaseibacillus rhamnosus strain HN001 (LactoB HN001™), which has been shown reduce postpartum depression, a milk fat globule membrane-enriched product, Lipid 70 (SurestartTM MFGM 70), implicated memory stress-susceptible Wistar Kyoto rats....

Appropriate intestinal barrier maturation during infancy largely depends on colonization with commensal bacteria. Faecalibacterium prausnitzii is an abundant obligate anaerobe that colonizes weaning and thought to maintain colonic health throughout life. We previously showed F. induced Toll-like receptor 2 (TLR2) activation, which linked enhanced tight junction formation. Therefore, we hypothesized enhances integrity, important factor in appropriate maturation. In order test metabolically...

The effect of two ovine-derived peptides on the morphology Staphylococcus aureus NCTC 4163 cells were investigated using transmission electron microscopy and atomic force microscopy. Both techniques showed that SMAP29, an α-helical peptide, induced cell lysis, whereas OaBac5mini, a proline/arginine-rich caused no observable morphological changes. This is consistent with previous experimental work which indicated SMAP29 death OaBac5mini acted by interacting inner cellular contents.

Abstract Lactobacilli are thought to be beneficial for human health, with lactobacilli-associated infections being confined immune-compromised individuals. However, Lactobacillus fermentum AGR1487 negatively affects barrier integrity in vitro so we hypothesized that it caused a pro-inflammatory response the host.
