The vacuolar protein sorting 35 (VPS35) is a major component of the retromer complex system, an ubiquitous multiprotein assembly responsible for and trafficking cargos out endosomes. VPS35 can regulate APP metabolism Aβ formation, its levels are reduced in Alzheimer's disease (AD) brains. We others demonstrated that genetic manipulation modulates phenotype mouse models AD. However, translational value this observation remains to be investigated.Triple transgenic mice were randomized receive...

Currently the entire human population is in midst of a global pandemic caused by SARS-CoV-2 (Severe Acute Respiratory Syndrome CoronaVirus 2). This highly pathogenic virus has to date >71 million infections and >1.6 deaths >180 countries. Several vaccines drugs are being studied as possible treatments or prophylactics this viral infection. M3CLpro (coronavirus main cysteine protease) promising drug target it significant role replication. Here we use X-ray crystal structure complex with...

Inflammatory bowel disease (IBD) encompasses several debilitating chronic gastrointestinal (GI) inflammatory disorders, including Crohn's and ulcerative colitis. In both conditions, mucosal inflammation is a key clinical presentation associated with altered serotonin (5-hydroxytryptamine or 5-HT) signaling. This 5-HT signaling also found across various animal models of Of the 14 known receptor subtypes, type 7 (5-HT

TAR DNA binding protein 43 (TDP-43) is a nucleic acid that associated with the pathology of cystic fibrosis and neurodegenerative diseases such as amyotrophic lateral sclerosis frontotemporal lobar dementia. We have developed robust, quantitative, nonradiometric high-throughput assay measuring oligonucleotide to TDP-43 using AlphaScreen® technology. Biotinylated single-stranded (bt-TAR-32) 6 TG repeats (bt-TG6) bound high affinity TDP-43, KD values 0.75 nM 0.63 nM, respectively. Both...

Cocaine use disorder currently lacks Food and Drug Administration–approved treatments. In rodents, the glutamate transporter-1 (GLT-1) is downregulated in nucleus accumbens after cocaine self-administration, increasing expression function of GLT-1 reduces reinstatement seeking. The β-lactam antibiotic ceftriaxone upregulates attenuates cue- cocaine-induced seeking without affecting motivation for natural rewards. Although shows promise treating disorder, it possesses characteristics...

Atrial fibrillation is the most prevalent form of cardiac arrhythmia. Current treatments extend atrial effective refractory period by nonselective blockade ion channels. An alternative approach selectively targeting Kv1.5 channel offers opportunity for therapeutic benefit with decreased risk adverse cardiovascular events. KVI-020 (4g) successfully demonstrated antiarrhythmic efficacy in a canine arrhythmia model, and these findings support its utility as an agent.

The dopamine D3 receptor is a prime target for developing treatments cocaine use disorders (CUDs). In this study, we conducted pre-clinical investigation of the therapeutic potential long-acting, partial agonist, MC-25-41. Male rats were pre-treated with MC-25-41 (vehicle, 1.0, 3.0, 5.6, or 10 mg/kg, intraperitoneal (IP)) five minutes prior to tests sucrose intake on either progressive ratio schedule reinforcement variable interval 60 s multiple consisting 4, 15-min components available in...
