Abstract NKCC and KCC transporters mediate coupled transport of Na + +K +Cl − K across the plasma membrane, thus regulating cell Cl concentration volume playing critical roles in transepithelial salt water neuronal excitability. The function these has been intensively studied, but a mechanistic understanding awaited structural studies transporters. Here, we present cryo-electron microscopy (cryo-EM) structures two cation-chloride cotransporters human NKCC1 (SLC12A2) mouse KCC2 (SLC12A5),...

The existence of a cilia-dependent cyst activation (CDCA) pathway underlying autosomal dominant polycystic kidney disease was identified by showing that progression following loss polycystins is significantly suppressed removal structurally intact cilia. We applied translating ribosome affinity purification RNASeq on pre-cystic mouse kidneys to determine cell-autonomous in vivo translatome associated with CDCA and Glis2 as an early effector polycystin signaling. Alzheimer's Disease (AD) manifests neuronal loss. On the premises of Grb2 overexpression in AD mouse brain and tissues patients, our study primarily focuses on stability cytoskeletal proteins context degenerative like conditions. Two predominant molecular features AD, extracellular accumulation Aβ oligomers intracellular elevation AICD levels, have been used to closely inspect series signaling events. In their presence, multiple pathways involving ROCK PAK1 lead disassembly cytoskeleton...

The ascertainment of elevated levels amyloid-β protein precursor intracellular domain (AICD) in Alzheimer's disease (AD) brains and the fact that it contributes to AD-like pathology has geared search toward a new paradigm. While studying endogenous as well overexpressed Grb2-AICD interact ion AD cell models, was found Grb2 co-localized compartments along with AICD. We report now these vesicles form clathrin dynamin independent manner. Both types mature into autophagosomes, merge lysosomes,...

SUMMARY The serotonin transporter (SERT) removes synaptic and is the target of anti-depressant drugs. SERT adopts three conformations: outward-open, occluded, inward-open. All known inhibitors outward-open state except ibogaine, which has an unusual profile stabilizes inward-open conformation. Unfortunately, ibogaine promiscuous cardiotoxic, limiting understanding ligands. We computationally docked over 200 million small molecules against stabilized SERT. Thirty-six top-ranking compounds...

Based on the observations that Grb2 overexpression altered trafficking route of amyloid-β protein precursor (AβPP) by inhibiting its release via exosomal vesicles, and subsequently increased endogenous level, in present study we aimed to elucidate mechanism traffic impairment role different domains this process. We found N-SH3 domain was involved vesicular localization. The C-SH3 could also form very small puncta, but were not characteristic containing vesicles. Vesicles N-SH3-SH2 had a...

Growth factor receptor-bound protein 2 (Grb2) is an adaptor which participates in trafficking pathways alongside its role signaling. Proteins important for actin remodeling and cellular compartmentalization contain SRC Homology 3 (SH3) binding motifs that interact with Grb2. While studying the Grb2-amyloid precursor (APP) intracellular domain (AICD) interaction Alzheimer's disease cell line models, it was seen Grb2 colocalized to compartments mature into autophagosomes. We present numerical results of the recently derived polaronic t-Jp model, which accounts for realistic long range Coulomb repulsion and strong Fröhlich electron-phonon interactions high temperature superconductivity in doped polar insulators. Different cluster geometries 18-sites with two particles are studied. It appears that ground state energy decreases hopping polarons indicating an increase tendency to couple each other. The entropy curves show system goes more ordered polaron all geometries.

The possibilities of the formation superlight small bipolaron in doped polar insulators have been discussed within t-Jp model derived from Fröhlich and realistic long range Coulomb interaction. Increase t/Jp dresses electrons forming polarons bipolarons. Formation inter-site (S1) bipolarons is favored by ratio. Entropy curves indicate that this lead to a more ordered state system.

We present numerical results of the t-J-V model in an 8-site tilted square cluster using exact diagonalization (ED) method with periodic boundary conditions. Effective hopping amplitude initially increases inter-site Coulomb repulsion (V), but decreases at larger V's. The hole-hole correlation distances smaller V. With increase repulsion, system becomes ordered. specific heat curves confirm non-Fermi liquid behavior under model.
