The unexpected, non-enzymatic S-glycosylation of cysteine residues in various proteins by per-O-acetylated monosaccharides is described. This artificial greatly compromises the specificity and validity metabolic glycan labeling living cells azido alkynyl sugars, which has been overlooked field for decades. It demonstrated that use unacetylated unnatural sugars can avoid artifact formation a corrected list O-GlcNAcylated O-GlcNAc sites HeLa assembled using N-azidoacetylgalactosamine (GalNAz).
Unnatural monosaccharides such as azidosugars that can be metabolically incorporated into cellular glycans are currently used a major tool for glycan imaging and glycoproteomic profiling. As common practice to enhance membrane permeability uptake, the unnatural sugars per-O-acetylated, which, however, induce long-overlooked side reaction, non-enzymatic S-glycosylation. Herein, we develop 1,3-di-esterified N-azidoacetylgalactosamine (GalNAz) next-generation chemical reporters metabolic...
Large-scale quantification of protein O-linked β-N-acetylglucosamine (O-GlcNAc) modification in a site-specific manner remains key challenge studying O-GlcNAc biology. Herein, we developed an isotope-tagged cleavable linker (isoTCL) strategy, which enabled isotopic labeling through bioorthogonal conjugation affinity tags. We demonstrated the application isoTCL mapping and O-GlcNAcylation sites HeLa cells. Furthermore, investigated sensitivity to sugar donor by quantifying levels under...
Immunoconjugates, including antibody-drug conjugates and Fc-conjugates, represent a rapidly growing class of therapeutics undergoing clinical development. Despite their popularity, the high intrinsic heterogeneity immunoconjugates often complicates development process limits widespread application. In particular, immunoconjugate charge variants exhibit markedly different colloidal stabilities, solubilities, pharmacokinetics, tissue distributions. Charge arise spontaneously due to degradation...
Abstract Chemoenzymatic modification of cell-surface glycan structures has emerged as a complementary approach to metabolic oligosaccharide engineering. Here, we identify Pasteurella multocida α2-3-sialyltransferase M144D mutant, Photobacterium damsela α2-6-sialyltransferase, and Helicobacter mustelae α1-2-fucosyltransferase, efficient tools for live-cell modification. Combining these enzymes with pylori α1-3-fucosyltransferase, develop host-cell-based assay probe glycan-mediated influenza A...
Phenolic acids and tanshinones are active principles in Salvia miltiorrhiza Bunge administered for cardiovascular cerebrovascular diseases. Jasmonic acid (JA) promotes secondary metabolite accumulation, but the regulatory mechanism is unknown S. miltiorrhiza. We identified characterized JA-responsive gene SmMYB97. Multiple sequence alignment phylogenetic tree analyses showed that SmMYB97 was clustered with AtMYB11, AtMYB12, ZmP1 subgroup S7 regulating flavonol biosynthesis. highly expressed...
O-linked N-acetylglucosamine (O-GlcNAc) glycosylation, a prevalent protein post-translational modification (PTM) that occurs intracellularly, has been shown to crosstalk with phosphorylation and ubiquitination. However, it is unclear whether interplays other PTMs. Here we studied its relationship ADP-ribosylation, which involves decorating target proteins the ADP-ribose moiety. We discovered poly(ADP-ribosyl)ation "eraser", glycohydrolase (PARG), O-GlcNAcylated at Ser26, in close proximity...
Proteins are ubiquitously modified with glycans of varied chemical structures through distinct glycosidic linkages, making the landscape protein glycosylation challenging to map. Profiling intact glycopeptides mass spectrometry (MS) has recently emerged as a powerful tool for revealing matched information sites and attached (i.e., glycosites), but is largely limited individual types. Herein, we describe Click-iG, which integrates metabolic labeling clickable unnatural sugars, an optimized MS...

O-linked β-N-acetylglucosaminylation (O-GlcNAcylation) is a ubiquitous post-translation modification occurring in both animals and plants. Thousands of proteins along with their O-GlcNAcylation sites have been identified various animal systems, yet the O-GlcNAcylated proteomes plants remain poorly understood. Here, we report large-scale profiling protein site-specific manner rice. We first established metabolic glycan labelling (MGL) strategy N-azidoacetylgalactosamine (GalNAz) rice...
Hypericum perforatum L. is a widely known medicinal herb used mostly as remedy for depression because it contains high levels of naphthodianthrones, phloroglucinols, alkaloids, and some other secondary metabolites. Quantitative real-time PCR (qRT-PCR) an optimized method the efficient reliable quantification gene expression studies. In general, reference genes are in qRT-PCR analysis their or suspected housekeeping roles. However, level cannot be assumed to remain stable under all possible...
Recently, some athletes were repetitively found to have rEPO positive results, including a characterized double-band pattern in blood samples, routine doping analysis. In contrast previous findings from excretion studies, this showed the same relative intensity even when samples collected weeks (/months) apart. We therefore suspected that these "positive" control related with novel pathway of endogenous EPO production. Thus, follow-up investigations warranted characterize origin such...
Abstract We explored the application of electron detachment dissociation (EDD) and infrared multiphoton (IRMPD) tandem mass spectrometry to fluorescently labeled sialylated oligosaccharides. Standard oligosaccharides a N ‐linked glycan released from human transferrin were investigated. EDD yielded extensive glycosidic cleavages cross‐ring in all cases studied, consistently providing complementary structural information compared with dissociation. Neutral losses satellite ions such as C−2H...
Twelve new hybrid compounds of Esculetin with nitric oxide (NO) donors and/or mitochondrial targeting groups were designed, synthesized, and evaluated for their anti-tumor activity mechanism in vitro vivo. Notably, the most potent compound A11 exhibited nanomolar antiproliferative on triple-negative breast cancer (TNBC) MDA-MB-231 cells (IC50 = 8 nM) a strikingly selective inhibitory effect. The involves cells' mitochondria, releasing high NO concentration, increasing expression cyclophilin...

Isosteviol is a beyerane tetracyclic diterpenoid with large variety of biological activities. In this article, series novel isosteviol derivatives containing the modification C-18 carboxyl group (5-12), C-16 carbonyl (14- 16) and heteroatom-containing frameworks fused structure (18-19) were synthesized evaluated for their in vivo acute antihyperglycemeric effects. Among them, compound 8 exhibited most potent Furthermore, primarily, structure-activity relationship (SAR) was also analyzed. The...
As a ubiquitous and essential posttranslational modification occurring in both plants animals, protein N-linked glycosylation regulates various important biological processes. Unlike the well-studied animal N-glycoproteomes, landscape of rice N-glycoproteome remains largely unexplored. Here, by developing chemical glycoproteomic strategy based on metabolic glycan labeling (MGL), we report comprehensive profiling seedlings. The seedlings are incubated with N-azidoacetylgalactosamine (GalNAz)...
